摘  要:目的研究辅酶Q致突变的可能性,探讨其是否具有遗传毒性。方法NIH种小鼠50只,按体重随机分为阴性对照组,阳性对照组,10.0g/kgBW,5.00g/kgBW和2.50g/kgBW辅酶Q组,每组10只,雌雄各半,通过微核试验观察小鼠经不同剂量的辅酶Q处理后,其骨髓细胞微核形成率的变化,探讨辅酶Q是否具有诱导小鼠骨髓细胞微核形成的作用;雄性NIH种小鼠25只,按体重随机分为阴性对照组,阳性对照组,10.0g/kgBW,5.00g/kgBW和2.50g/kgBW辅酶Q组,通过小鼠精子畸形试验观察小鼠经不同剂量的辅酶Q处理后,其精子畸形率的变化,探讨辅酶Q是否具有诱导小鼠精子畸形形成的作用;采用Ames试验,观察经5000,1000,200,40和8g/皿的辅酶Q处理的鼠伤寒沙门氏菌TA97,TA98,TA100和TA102四株试验菌株回复突变情况。分析辅酶Q是否具有诱导鼠伤寒沙门氏菌TA97,TA98,TA100,TA102四株试验菌株产生回复突变的作用。结果2.5~10.0g/kgBW辅酶Q处理的各剂量组小鼠的骨髓细胞微核形成率未出现明显增加,与对照组相比差异无统计学显著性意义(P〉0.05);2.5~10.0g/kgBW辅酶Q处理的各剂量组小鼠的精子畸形率未出现明显增加;与对照组相比差异无统计学显著性意义(P〉0.05);在加和不加代谢活化系统条件下,8~5000g/皿辅酶Q处理的各剂量组的鼠伤寒沙门氏菌TA97,TA98,TA100,TA102四株试验菌株回复突变率均未出现明显升高,与相应的对照组相比差异无统计学显著性意义(P〉0.05)。结论在该研究条件下未发现辅酶Q具有直接或间接的致突变作用,也未发现其具有遗传毒性。Objective To study the possibility of mutagenie and the genetic toxicity on the coenzyrne Q. Methods With the treatment of different doses of the eoenzyme Q, the effect of the mouse bone marrow ceils micronueleus and sperm abnormalities was observed by micronucleus test and sperm shape abnormality test. Through Ames test,the reverse mutation of four strains Salmonella typhimurium (TA97,TA98, TA100, TA102) was observed, with the treatment of different doses of the coenzyme Q. Results The bone marrow cells micronucleus rate and sperm deformity rate of mice treated with 2.5-10. 0 g/ kg BW the coenzyme Q was not significant increase, there were not significant difference compared with the corresponding eontrol group (P2〉0.05). Under the conditions of with and without metabolic activation system, the reverse mutation rate of four strains Salmonella typhimurium (TA97, TA98, TA100, TA102) was not significant increase up to 5 000 ug/plate,compared with the control group,no significant difference (P〉0. 05). Conclusion It was not found the direct or indirect mutagenic effect and the genetic toxicity associate with the coenzyme Q.
