摘  要:目的:探讨即刻早期反应基因-1(immediate early response gene X-1,IEX-1)转染人卵巢癌细胞SKOV3后,对顺铂药物敏感性的影响。方法:提取并鉴定重组质粒pEGFP-N1-IEX-1,采用脂质体介导将重组质粒转染到人卵巢癌细胞SKOV3,RT-PCR检测转染前后IEX-1mRNA和蛋白的表达情况,采用四甲基偶氮唑蓝(MTT)法检测转染后卵巢癌细胞SKOV3增殖情况以及对顺铂敏感性的变化。结果:成功转染IEX-1基因后可检测到IEX-1mRNA的高表达;转染组细胞的增殖速度增快(P<0.05),但对顺铂的敏感性明显增强(P<0.05)。结论:IEX-1可增强卵巢癌细胞的体外增殖能力,并增强卵巢癌细胞体外对顺铂化疗的敏感性,为肿瘤的有效干预和治疗提供新的靶点。Objective:To explore the recombined plasmid pEGFP-N1-IEX-1 transfection and its influence on the sensitivity to cisplatin on human ovarian cell line SKOV3. Methods:The recombined plasmid pEGFP-N1-IEX-1 was purified and verified by PCR.The expression of IEX-1 mRNA was examined by RT-PCR.MTT was used to detect the effects of IEX-1 gene transfection on proliferation and sensitivity to cisplatin of ovarian cancer cell line SKOV3. Results:There was low expression of IEX-1 mRNA in SKOV3 cells and high expression of IEX-1 mRNA was detected after transfection.After transfection,the MTT results showed that cell proliferation rate was much faster(P<0.05) and more sensitivity to cisplatin compared with the normal group(P<0.05). Conclusion:IEX-1 can promote the proliferation ability and enhance the sensitivity to cisplatin of ovarian cancer cell line SKOV3 in vitro,which provides new targets for effective intervention and treatment of ovarian cancer.
