摘 要:目的:采用反相高效液相色谱法同时测定山核桃叶中汉黄芩素、白杨素、小豆蔻明、球松素查尔酮、松属素的含量。方法:采用戴安Ultimate 3000系统,使用戴安Acclaim 120 C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.1%乙酸水溶液(70∶30)为流动相,流速1.0 mL.min-1,柱温为30℃,检测器为戴安Ultimate 3000 DAD,以对应化合物最大吸收波长(汉黄芩素272nm、白杨素262 nm、小豆蔻明342 nm、球松素查尔酮342 nm、松属素284 nm)为检测波长,同时对山核桃叶中5个黄酮苷元进行含量测定。结果:在上述条件下,汉黄芩素、白杨素、小豆蔻明、球松素查尔酮、松属素浓度分别在1.27~25.3、2.56~51.2、1.29~25.6、5.01~100.2和5.06~101.2μg.mL-1范围内线性关系良好,平均加样回收率(n=9)分别为98.83%、99.27%、101.1%、98.70%和99.54%;测得山核桃叶中上述的5个化合物平均含量(n=3)分别为0.0183%、0.1142%、0.0618%、0.7934%和0.3368%。结论:本法同时测定山核桃叶中汉黄芩素、白杨素、小豆蔻明、球松素查尔酮、松属素5个黄酮苷元,为评价和监控山核桃叶质量及其深入研究与开发奠定了良好基础。Objective: Using the RP - HPLC method for simultaneous determination of five flavonoid aglycones ( wogonin, chrysin, cardamomin, pinostrobin chalcone, and pinocembrin) in the leaves of Carya cathayensis Sarg.Methods: The DIONEX Ultimate 3000 HPLC system coupled with DAD was carried out, and the Acclaim 120 C18 column (250 mm×4.6 mm,5μm) was used with the mobile phase consisting of methanol and 0. 1% acetic acidaqueous solution (70: 30) to determine the content of the five flavonoid aglycons. The flowrate was 1.0 mL · min^- 1, the column temperature was at 30 ℃, The ultimate 3000 DAD detector was used. Consistent with the maximum absorption wavelength of the compound, the detection wavelengths were 272 nm( wogonin), 262 nm ( chry-sin),342 nm (cardamomin and pinostrobin chalcone), and 284 mn (pinocembrin). Results: The calibrationcurves of the five flavonoid aglycons were linear in the range of 1.27 - 25.3,2.56 - 51.2,1.29 - 25.6,5.01 - 100.2 and 5.06 - 101.2 μg·mL^-1 and the average recoveries (n =9) were 98.83%,99.27% 101.1%98.70% , and 99.54% for wogonin, chrysin, cardamomin, pinostrobin chalcone, and pinocembrin, respectively. Additionally,the average contents of the 5 compounds mentioned above were 0. 0183%, 0. 1142%, 0. 0618%,0. 7934% ,and 0. 3368% ,respectively. Concluslon: The method can simultaneously determine the content of the five flavonoid aglycones in the leaves of C. cathayensis ,which paves the way for evaluation, quality control and further exploitation of the leaves of C. cathayensis.
