摘 要:在真核生物中DNA甲基化是一种重要的表观遗传学修饰方法,其对基因的表达调控起到非常重要的作用。本研究以高肌间脂肪含量猪地方品种莱芜猪与大白猪的正反杂交后代为研究对象,利用F-MSAP技术,应用筛选的16对引物扩增,检测其肌肉组织的全甲基化程度,揭示不同品种肌肉组织间基因组全甲基化水平差异。结果表明以莱芜猪为父本,大白猪为母本的正交一代找到2 031个甲基化位点,以大白猪为母本,莱芜猪为父本的反交一代检测到1 905个甲基化位点,总甲基化水平分别为57.20%、53.5%。并且反交一代半甲基化水平显著高于正交一代(P<0.05),正交一代全甲基化水平极显著高于反交一代(P<0.01)。DNA methylation is one of the main epigenetic modification mechanisms in eukaryotic organisms,playing a crucial role in the regulation of gene expression. In this study,we put F1 hy-brid with Laiwu pigs and Yorkshine as research object, used the fluorescence-labeled methylation-sensitive amplified polymorphism (F-MSAP) method and used 16 pairs of selective primers to assess the extent and pattern of cytosine methylation in the muscle. The incidence of DNA methylation was approximately 57.20% in the muscle of direct cross and total of 2 031 sites were found to be methylated at cytosines. 53.50% in the reciprocal cross and total of 1 905 sites were found to be methylated at cytosines. As type II of methylation status,the reciprocal cross was higher than the direct cross significantly(P〈0.05). As type III of methylation status, the direct cross was higher than the reciprocal cross exthreme significantly(P〈0.01).
