重组ING4基因对卵巢癌OVCAR细胞生长抑制作用的实验研究

摘　　要：目的:观察ING4基因转染人卵巢癌OVCAR细胞后对细胞的生长抑制效应。方法:采用ING4的重组腺病毒载体(Ad-ING4)感染人卵巢癌细胞株OVCAR,用RT-PCR法检测ING4在OVCAR细胞中的表达;MTT法和流式细胞技术检测ING4基因表达对OVCAR细胞的生长抑制和凋亡效应;Western印迹法检测ING4对OVCAR细胞中bax、bcl-2、Ki67表达的影响。结果:在OVCAR细胞中ING4基因的表达对OVCAR细胞增殖有明显抑制作用,并可诱导细胞凋亡;Western印迹法检测结果提示卵巢癌组织中ING4基因表达下降的同时,Ki67表达下调,Bax表达上调,Bcl-2的表达下调。结论:转染ING4基因可抑制OVCAR人卵巢癌细胞的增殖,可通过改变Bax,Bcl-2等凋亡相关基因的表达诱导细胞凋亡。Objective： To observe inhibitory effect on cell growth after the ING4 gene transfeetion of human ovarian cancer cell line OVCAR. Methods： ING - 4 the recombinant adenoviruses sub （ Ad - ING4 ） infection in humanovarian cancer cell line cell line OVCAR, detected by RT - PCR method for ING4 expression in OVCAR cells. MTr assay and flow cytometry were used to detect ING4 gene expression OVCAR cell growth inhibition and apoptotic effect. To detect expression of bax, bcl - 2, Ki67 in OVCAR cells by Western blotting method. Results： ING4 gene expression in OVCAR cells OVCAR Cell proliferation was significantly inhibited, and induce apoptosis. Western blotting method prompted that ING4 gene expression in ovarian cancer tissue decreased while downregulation of Ki67, upregulation of Bax, downregulation of Bcl -2. Conclusion：The ING4 gene transfection can inhibit the proliferation of OVCAR human ovarian cancer and throw the change of Bax, Bcl -2 genes induced apoptosis.
