摘 要:目的:构建骨肉瘤的耐药细胞模型,探讨IER3基因在骨肉瘤耐药中的表达情况及意义。方法:采用递增药物浓度冲击诱导法构建耐阿霉素人骨肉瘤细胞模型(MG-63/ADM),应用RT-PCR和Western blot检测IER3在敏感细胞和耐药细胞中的基因和蛋白水平表达情况。结果:经过6个月的诱导,建立了对于不同阿霉素浓度耐药的细胞模型分别标记为MG-63/ADM0.1、MG-63/ADM0.4、MG-63/ADM1.0、MG-63/ADM4.0,细胞形态在光镜下明显改变,MG-63/ADM细胞中IER3的表达量明显高于MG-63。结论:药物冲击诱导可以使骨肉瘤细胞产生对于化疗药物的耐药性,为研究骨肉瘤耐药性及其逆转提供实验基础,IER3可能参与了骨肉瘤对于阿霉素的耐药机制。Objective:Constructing osteosarcoma resistance cell model, discussing the expression and significance of IER3 gene in osteosarcoma of drug resistance. Methods:To construct MG -63/AI)M by increasing the concentration of ADM impacting and inducing, to detect IER3 gene and protein expression in two kinds of cells by RT - PCR and western blot. Results:After six months of induction, we constructed cell models of resistance to different concentration of ADM, marked MG - 63/ADM0. 1, MG - 63/ADM0.4, MG - 63/ADM1.0, MG - 63/ADM4.0. Cell mor- phology in the pathologic change obviously, IER3 in MG -63/ADM expression quantity was obviously higher than that of the MG -63. Conclusion:Drugs shock induced can make osteosarcoma cells to produce resistance to chemotherapy drugs, the research provides experimental basis for osteosarcoma resistance and reversion, IER3 may be involved in the mechanism of drug resistance of osteosarcoma.
