印甲薪对人乳腺癌细胞株MCF-7体外侵袭能力的作用研究

摘　　要：目的:研究非甾体类抗炎药印甲薪对人乳腺癌细胞株MCF-7体外侵袭能力的影响。方法:通过细胞增殖抑制实验确定可应用于后续实验的印甲薪药物浓度,应用Transwell细胞体外侵袭实验研究印甲薪对MCF-7细胞移行及浸润能力的影响。结果:在安全浓度范围内,随着印甲薪浓度的增加,MCF-7细胞的移行能力无明显变化,但浸润能力明显下降,与对照组相比有明显的统计学意义(P<0.05),并且浸润能力与印甲薪浓度呈负相关(r=-0.985)。结论:印甲薪可以明显抑制MCF-7细胞的体外侵袭能力,并且该抑制作用呈现浓度依赖性。Objective：To study the effect on invasion ability of breast cancer cell line MCF -7 by indomethacin in vitro. Methods： Cell growth inhibition assay was performed to determine the non - cytotoxic dose range of indomethacin for the next experiment, then Transwe11 migration assay and matrigel infiltration assay were performed to assess the effect of indomethacin on the migration and infihration ability of MCF - 7. Results： The migration ability of MCF - 7 cells was not affected by indomethacin in the range of safe dose, and the invasion ability was significantly inhibited in a dose dependent manner（ r = -0. 985 ）, there was significant difference between experiment and control group（ P 〈0. 05）. Conclusion：The non- cytotoxic level of indomethacin can inhibit the invasion of MCF -7 cells in vitro, and the inhibition effect is related to the concentration of indomethacin.
