摘 要:目的研究高压氧预适应(hyperbaric oxygen preconditioning,HBOP)对小鼠大脑中动脉闭塞(middle cerebralartery occlusion,MCAO)的保护作用及对其脑内P2X7受体(P2X7R)表达的影响。方法将72只雄性C57BL/6小鼠分为:假手术组(sham)、高压氧预适应假手术组(HBOP+sham)、单纯MCAO组(MCAO)、高压氧预适应MCAO组(HBOP+MCAO)。通过线栓法建立小鼠MCAO模型,术后24 h采用Longa法对小鼠进行神经功能学评分,氯化三苯基四氮唑(TTC)染色法判断脑梗死情况,TUNEL法观察神经细胞凋亡情况,实时荧光定量PCR(RT-qPCR)法观察P2X7受体mRNA的表达。结果 MCAO建模后24 h,HBOP+MCAO组神经功能学评分明显低于MCAO组(2.33±0.44 vs 2.89±0.59,P<0.05);HBOP+MCAO组脑梗死范围明显小于MCAO组[(44.08±8.46)%vs(63.00±5.25)%,P<0.05];HBOP+MCAO组凋亡阳性细胞数和P2X7R mRNA表达水平也明显低于MCAO组(P<0.05,P<0.01)。结论高压氧预适应可以减轻实验性MCAO小鼠的神经功能学损害,缩小梗死灶体积,减少细胞凋亡,具有明确的脑保护作用,其保护作用可能与延缓MCAO后P2X7R的表达升高有关。Objective To study the neuroprotective effect of hyperbaric oxygen preconditioning(HBOP), and the effect of HBOP on the expression of P2X7 receptor in mouse middle cerebral artery occlusion(MCAO). Methods Total 72 C57BL/6 mice were divided into 4 groups: sham-operated group, HBOP+sham-operated group, MCAO group and HBOP+MCAO group. The MCAO model was established by suture occluded method. The neurological deficits and infarct volumes were detected at 24 h after MCAO in mice. The apoptotic cells were detected by using the terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL). The expression of P2X7 receptor mRNA was determined by RT-QPCR. Results Compared with MCAO group, the scores of neurological deficits were reduced obviously in HBOP+MCAO group(P〈0.05); compared with the MCAO group, the rate of infarct volumes decreased significantly in HBOP+MCAO group( P〈0.05); compared with MCAO group, the apoptotic cells were significantly diluted in HBOP+MCAO group(P〈0.05); compared with MCAO group, the expression of P2X7 receptor mRNA was reduced in HBOP+MCAO group(P〈0.01). Conclusion The results suggest that the HBOP have a absolutely neuroprotective effect, it can attenuate the injury of the experimental MCAO model, reduce the infarct volume and dilute the apoptosis, and the neuroprotective effect maybe concerned with the delayed of P2X7 receptor’s expression after MCAO.
