胰腺癌组织中Her-2/neu的表达与临床病理因素的关系

摘　　要：目的检测Her-2/neu在胰腺癌中的表达,探讨其在胰腺癌中发生发展的作用。方法收集50例原发性胰腺癌组织标本和36例癌旁组织,采用免疫组织化学法、逆转录荧光定量PCR法(RT-QPCR)和蛋白免疫印迹(Westernbolt)检测胰腺癌及癌旁胰腺组织中Her-2/neu的表达;并分析Her-2/neu表达与胰腺癌病理参数的关系。结果 Her-2/neu在胰腺癌组织中表达明显高于癌旁组织(P<0.05);Her-2/neu的表达与胰腺癌的病理组织分级、肿瘤转移、临床分期和1年期生存率关系密切,组织学Ⅲ级中表达明显高于Ⅰ、Ⅱ级(P<0.05),Ⅲ～Ⅳ期表达明显高于Ⅰ～Ⅱ期(P<0.05);有转移组明显高于无转移组(P<0.05);低于1年期生存患者表达明显高于1年期的表达(P<0.05)。Her-2/neu的表达与患者的年龄、性别、发病部位、肿瘤增殖活性无关。结论 Her-2/neu高表达与胰腺癌的发生发展有关。Objective To investigate the expression of Her-2/neu in pancreatic carcinoma（PC） and to explore its role in carcinogenesis and progress of PC.Methods 50 specimens of primary PC and 36 adjacent tissue specimens were collected.The expressions of Her-2/neu were detected by immunohistochemistry,RT-QPCR and Western Blot.The correlation between the expressions of Her-2/neu and the biological parameters of PC was analyzed.Results The expression of Her-2/neu in PC tissues was significantly higher than that in PC adjacent tissues（P〈0.05）.The expression of Her-2/neu was closely correlated with PC＇s pathological classification,tumor metastasis,clinical stage and 1-year survival rate,which at histology Ⅲ grade was significantly higher than that at Ⅰ,Ⅱ grade（P〈0.05）,and its expression at Ⅲ～Ⅳ stage was significantly higher than that atⅠ～Ⅱ stage（P〈0.05）.The expression of Her-2/neu in patients with tumor metastasis was significantly higher than that in patients without tumor metastasis（P〈0.05）,and its expression in patients under 1-year survival life was significantly higher than that of patients over 1-year survival life（P〈0.05）.However the expression of Her 2/neu was not related to patients＇ age,sex,lesion position and proliferation activity of the carcinoma.Conclusion The overexpression of Her-2/neu is related to pathogenesis and progress of pancreatic carcinoma.
