摘  要:目的:探讨过氧化物酶体增殖激活受体γ(peroxidase body proliferation activated receptorγ,PPARγ)在不同分子亚型乳腺癌中的表达及其与乳腺癌预后的关系。方法:采用免疫组织化学法检测120例乳腺癌组织和20例乳腺良性肿瘤组织中PPARγ的表达。采用蛋白印迹法(western blotting)和实时荧光定量逆转录-聚合酶链反应(RT-PCR)法检测正常乳腺上皮细胞株和具有不同转移潜能的乳腺癌细胞株中PPARγ蛋白和mRNA表达的情况,分析PPARγ与患者临床预后的关系。结果:在不同分子亚型的乳腺癌组织中,PPARγ蛋白在三阴性乳腺癌(triple-negative breast cancer,TNBC)癌组织中表达最低(P<0.05);在不同分子亚型的乳腺癌细胞株中,PPARγmRNA和PPARγ蛋白在TNBC细胞株MDA-MB-231中表达最低;PPARγ在良性乳腺疾病、TNM I期乳腺癌和TNM III期乳腺癌中表达依次降低;PPARγ的表达与乳腺癌组织学分级、TNM分期、淋巴结转移、肿瘤直径和Ki-67呈显著负相关(P<0.05);PPARγ的高表达与患者总体生存率呈正相关(P<0.05)。结论:PPARγ的高表达是乳腺癌患者预后良好的指标。Objective:To study the expression levels of PPARγ in different molecular subtypes of cell lines and tissues of hu man breast cancer and its correlation with prognosis. Methods:The expression levels of PPARγ protein were detected by im munohistochemistry in 20 benign breast tumor tissue samples and 120 breast cancer tissue samples. Western blotting and real time polymerase chain reaction were used to detect PPARγ protein and mRNA expression levels in normal mammary gland epi- thelial cell line and breast cancer cell lines with different metastatic potential. The relationship between PPARγ expression and breast cancer prognosis was also analyzed. Results: The expression level of PPAR 3' protein was the lowest in triple negative breast cancer tissues(P〈0. 05). Among all breast cancer cell lines, PPAR γ mRNA and protein levels were the lowest in triple negative breast cancer cell line MDA MB-231. The expression levels of PPAR γ in benign breast diseases, TNM stage I breast cancer and TNM stage III breast cancer were reduced in turn. The PPAR γ expression level was negatively correlated with breast cancer histologic grading, TNM staging, lymph node metastasis, tumor size and ki-67(P〈0.05). High expression of PPAR y was positively correlated with the overall survival rate of patients (P〈.0.05). Conclusions: The expression level of PPAR γ is an independent prognostic factor in breast cancer.
