摘  要:目的:比较血管内皮生长因子(vascular endothelial growth factor,VEGF)mRNA在体外培养的成人虹膜色素上皮(irispigment epithelium,IPE)细胞与视网膜色素上皮(retinal pigment epithelium,RPE)细胞中的表达情况,探讨IPE细胞移植替代RPE细胞移植治疗年龄相关性黄斑变性(age related macular degeneration,ARMD)的可行性。方法:取生长近融合的传代2次的IPE及RPE细胞,Trizol一步法提取总RNA,采用逆转录聚合酶链反应(RT-PCR)方法检测VEGF mRNA的表达水平。结果:体外培养的IPE与RPE细胞均可表达VEGF mRNA,IPE细胞的VEGF mRNA表达量较RPE细胞低,差异有统计学意义(P<0.01)。结论:以自体IPE细胞替代RPE细胞进行移植,可能对于ARMD等新生血管增殖性疾病的治疗是有益的。Objective:To study the mRNA expression of VEGF in iris pigment epithelial cells (IPE cells) and retinal pigment epithelial cells (RPE cells) cultured in vitro, and to investigate the feasibility of the transplantation of autologous IPE cells to substitute RPE cells in the treatment of age related macular degeneration (ARMD). Methods:Total RNA was extracted from cultured adult human IPE cells and RPE cells of second passage at near confluence by using Trizol reagent. Semi-quantitative reverse transcription polymerase chain reaction (RT PCR) was conducted to detect the expression of VEGF at mRNA level. Resuits: VEGF mRNA expression was detected in both IPE cells and RPE cells. The expression level of VEGF mRNA in IPE cells was significantly lower compared with that in RPE cells (P〈0.01). Conclusions: Transplantation of 1PE cells may he better than transplantation of RPE cells in the treatment of ARMD.
