摘  要:目的探讨吉非替尼(Gefitinib)与染料木黄酮(Genistein)联合应用对人非小细胞肺癌(NSCLC)细胞株H1975增殖与凋亡的影响。方法采用MTT比色法观察两种药物单用及联用对人非小细胞肺癌细胞株H1975细胞增殖的影响;流式细胞仪检测细胞凋亡率;Western blotting检测凋亡相关基因PARP和caspase-3的蛋白水平。结果Gefitinib与Genistein联合用药对H1975细胞的抗增殖效应优于Gefitinib单药组,协同增效作用明显,抑制率升高,并呈现剂量依赖的抗增殖效应,差异有统计学意义(P<0.05);两药联用细胞凋亡率为(68.70±7.13)%,与正常对照组的(3.12±0.75)%、Genistein组的(35.95±2.32)%、Gefitinib组的(42.03±4.38)%比较,差异有统计学意义(P<0.05);与单药组比较,两药联用组PARP和caspase-3的断裂片段水平增加。结论 Genistein可以增强Gefitinib对H1975细胞的增殖抑制作用并促进肿瘤细胞的凋亡。Objective To investigate the effect of Gefitinib combined with Genistein on the proliferation and apoptosis of non small cell lung cancer (NSCLC) cell H1975. Methods The anti-proliferation effect of Gefitinib or/and Genistein on NSCLC cells H1975 was studied by MTF assay; apoptosis rate was detected by flow cytometry; the protein level of PARP and easpase-3 were assayed by Western blotting. Results Gefitinib combining with Genistein showed more obviously antiproliferative effects on H1975 cells than Gefitinib alone. The inhibition rate increased significantly and showed a dose-dependent effect(P〈O.05). Gefitinib synergized with Genistein could significantly enhaneed apoptosis and showing an apoptosis rate of (68.70±7.13)%, while the apoptosis rates of control group, Genistein group, and Gefitinib group were (3.12±0.75)%, (35.95±2.32)%, and (42.03±4.38)%, respectively(P〈0.05). Compared with Gefitinib group, the fracture fragments of PARP and caspase-3 in combination group were up regulated in protein level.
