摘 要:目的观察核因子-κB(NF-κB)抑制剂吡咯烷二硫氨基甲酸盐(PDTC)对毛果芸香碱诱导癫痫大鼠梨状皮层的保护性作用。方法将26只正常雄性SD大鼠随机分为3组:正常对照组(NS组,n=6)、毛果芸香碱诱导癫痫组(SE组,n=10)和PDTC干预组(PDTC组,n=10)。SE组一次性腹腔注射毛果芸香碱诱导大鼠癫痫发作;PDTC干预组分别于造模前24h、20min、SE发作后24 h腹腔注射PDTC 100mg.kg-1;NS组给予等量生理盐水。于造模成功后48h处死各组大鼠,采用Nissl染色和Fluoro-Jade C(FJC)染色法分别检测并比较各组大鼠梨状皮层存活神经细胞情况和退行性神经元数目。结果 Nissl染色结果显示,SE组大鼠梨状皮层大量神经细胞丢失,胞体结构完整性遭到破坏。FJG染色结果显示,SE组FJC阳性细胞数目明显增多;经PDTC干预后可明显改善梨状皮层神经损伤,FJC阳性细胞数明显减少,差异有统计学意义(P<0.05)。结论 PDTC对毛果芸香碱诱导癫痫大鼠梨状皮层损伤具有明显的保护性作用。Objective To investigate the protection effect of the selective nuclear factor-KB(NF-KB) inhibitor-pyrroli- dine dithiocarbamate (PDTC) on the pifiform cortex (PC) of pilocarpine-induced status epileptic (SE) rats. Methods Twen- ty-six male SD rats were randomly divided into three groups : normal control group ( NS group, n = 6 ), pilocarpine-induced SE group( SE group, n = 10), and PDTC intervention group( PDTC group, n = 10). The rats in SE group received pilocarpine only once at a high dose(320 mg · kg^-1 ) ;the PDTC group received PDTC( 100 mg · kg^-1 ) 24 hours,20 rain before induced-SE as well as SE 24 hours later respectively ; the NS group accepted the same volume of saline. Then all rats were sacrificed 48 hours after SE to study the surviving and the degenerating neurons in the PC of the three different groups using Nissl and Fluoro Jade C(FJC) staining methods. Results Nissl staining indicates that SE resulted in massive neuron loss in the PC of the rats, at the same time there were large number of FJC positive degenerative cells;pretreatment with PDTC significantly alleviated the neuronal damage in the PC and markedly reduced the FJC positive cells. Conclusion PDTC can significantly protect the PC of the pilocarpine-induced SE rats.
