摘  要:目的建立以聚β-氨基酯为载体的格列吡嗪纳米粒制备方法及其药物含量的高效液相色谱测定方法。方法分别采用改良复乳-溶剂挥发法和改良自乳化-溶剂扩散法制备纳米粒,比较其性质差异;高效液相色谱条件为:Easy Guard预柱(10mm×4.6mm),Ultimate xb-C18柱(250mm×4.6mm,5μm);流动相:0.1mol.L-1 NaH2PO4(用2.0mol.L-1 NaOH调pH为6.00±0.05)-甲醇(55∶45);流速:1.0mL.min-1;检测波长:225nm;进样量:20μL;柱温:35℃。结果改良复乳-溶剂挥发法制备纳米粒粒径较改良自乳化-溶剂扩散法粒径小;格列吡嗪在0.14~98.88mg.L-1范围内质量浓度与峰面积线性关系良好(r=0.999 7),平均回收率为100.03%,平均RSD为1.34%。结论采用改良复乳-溶剂挥发法制备纳米粒较佳;文中建立的HPLC法专属性好,精密度好,回收率高,可准确测定纳米粒中格列吡嗪的含量。Objective To establish a preparation method of glipizide nanoparticles with the carrier of poly (fl-amino esters), and to de- velop an HPLC method for the determination of glipizide. Methods The nanoparticles were prepared by using a modified double e- mulsion-solvent evaporation method and an improved self-emulsification-solvent diffusion method. The Easy Guard precolumn ( 10 mm×4.6 mm) ,and ultimate xb-C18 column (250 mm〉(4.6 mm,5 μm) were used with 0. 1 mol · L ^-1 sodium dihydrogen phos- phate (adjusted pH with 2.0 mol · L-1 sodium hydroxide to 6.00+0.05)-methanol (55: 45) as the mobile phase. The flow rate was 1.0 mL · min-x and the wavelength of detection was set at 225 nm,the sample loaded was 20 μL and the column temperature was 35 ℃. Results The nanoparticles particle size prepared by the modified double emulsion-solvent evaporation method was smal- ler than that by the improved self-emulsification-solvent diffusion method. A good linear relationship was found between the con- centration of glipizide and the peak area in the range of 0.14-98.88 mg·t-1 (r=0. 9997), the average recovery was 100.03 %, and average RSD was 1.34%. Conclusions It was a better way to use the modified double emulsion-solvent evaporation method to pre- pare the nanoparticles,and the developed HPLC method is reliable and accurate to determine the content of glipizide.
