摘  要:目的:总结PUMA蛋白的结构与功能、促凋亡及其转录调控的作用机制,介绍PUMA蛋白参与线粒体自噬、翻译后磷酸化调控及与之相关的信号通路等最新研究进展。方法:应用PubMed及CNKI期刊等全文数据库检索系统,以"p53上调凋亡调制物(PUMA)、凋亡和肿瘤"等为关键词,检索2002-2012年的相关文献。共检索到英文文献108篇,中文文献12篇。纳入标准:1)PUMA的基本特性;2)PUMA的促凋亡机制;3)PUMA的调控,包括转录调控和翻译后调控;4)PUMA的信号通路及调节因子。根据纳入标准符合分析的文献27篇。结果:PUMA通过BH3结构域的绑定功能发挥促凋亡作用及参与线粒体自噬;PUMA的调控机制主要有转录水平的调控(p53依赖/非依赖途径)及新发现的翻译后磷酸化调控机制;一系列PUMA新的调节因子和信号通路的发现,可以为治疗肿瘤的新抗癌药物提供依据。结论:通过对PUMA蛋白的结构与功能及其调控机制的进一步研究,将为人类肿瘤的诊断和治疗提供新的途径与方法。OBJECTIVE: To summarize the current articles about PUMA protein structural characteristics, function, pro apoptotic role and transcriptional regulation mechanisms of PUMA,and introduce the latest research of PUMA in the mitophagy,Post translational mechanisms and some novel signal pathways. METHODS: By PubMed and CNKI retrieval system,with "PUMA,apoptosis,tumour" as the key words,a total of 108 papers of literatures in English and 12 in Chi- nese were retrieved from 2002 to 2012. Choice criteria:l) The basic characteristics of PUMA 2) PUMA pro-apoptotic mechanisms; 3)PUMA regulation,including PUMA transcriptional regulation and Post-translational mechanisms; 4)PU- MA signal pathways and its regulatory factors. The articles that were repetitive studied and had no relations with this study were excluded,and 27 articles were included in reference study finally. RESULTS: The binding function of puma,in which depended the BH3 domain play pro-apoptotic role and take part in mitophagy. PUMA protein is subject to two ways of regulation:both through transcriptional (p53 dependent/independent) and a novel post-translational mechanisms. The research results of its some novel regulatory factors and signal pathways will provide the basis for development of an- ti-cancer drug in treating tumors. CONCLUSION: The further research of PUMA protein structure, function and its regu- lation mechanisms will provide the new ways for the diagnosis and treatment of human tumors.
