摘  要:目的比较马钱子超微粉与粗粉对小鼠毒性质反应量效关系的差异。方法采用急性毒性试验与连续给药7天的重复给药毒性试验,以死亡率为质反应阳性数,以半数致死剂量(LD50)、最小致死剂量(MLDLD1)、最大耐受剂量(MTDLD0)为考察指标,观察微粉化对制马钱子质反应量效、时效关系的影响。结果马钱子超微粉致雄鼠的LD50、MLDLD1、MTDLD0依次为194.7、100、85mg/kg,致雌鼠的依次为163.0、85、70mg/kg;马钱子粗粉致雄鼠的LD50、MLDLD1、MTDLD0依次为295.7、115、100mg/kg,致雌鼠的依次为271.8、100、85mg/kg。随着剂量的增加,马钱子超微粉较粗粉致小鼠死亡潜伏期缩短更快。结论马钱子超微粉及粗粉对小鼠的毒性反应相似,其急性毒性存在剂量依赖性的时反应量效关系,微粉化后其致死质反应急性毒性与重复给药毒性明显增强。Objective To compare the differences of dose-effect relationship in the toxicity qualitative response between the ultrafine powder and coarse powder of strychni semen on mice.Methods The acute toxicity test and 7-day repeated dose toxicity test were used.The dose-effect and time-effect of ultrafine powder on the qualitative response of strychni semen were observed according to the mortality rate,median lethal dose(LD50),minimum lethal dose(MLDLD1)and maximum tolerated dose(MTDLD0).Results The LD50,MLDLD1and MTDLD0of strychni semen ultrafine powder on male mice was 194.7mg/kg,100mg/kg and 85mg/kg respectively and on female mice was 163.0mg/kg,85mg/kg and 70mg/kg respectively.The LD50,MLDLD1and MTDLD0of strychni semen coarse powder on male mice was 295.7mg/kg,115mg/kg and 100mg/kg respectivelyand on female mice was 271.8mg/kg,100mg/kg and 85mg/kg respectively.As the dose increased,the death latency of mice induced by strychni semen ultrafine powder was shortened more quickly than coarse powder.Conclusion The toxicity response of strychni semen ultrafine powder and coarse powder is similar in mice.The acute toxicity has a time-effect relationship with dosedependence.The micronization can increase the acute toxicity and repeated dose toxicity in the lethal qualitative response.
