摘  要:目的观察氧化苦参碱对膀胱癌T24细胞增殖及凋亡的影响,并初步探讨其作用机制。方法不同浓度(0.625、1.25、2.5 mg/mL)氧化苦参碱作用于膀胱癌T24细胞,采用流式细胞术、光镜及电镜观察其诱导膀胱癌T24细胞的凋亡作用。用RT-PCR检测细胞凋亡相关基因survivin及caspase-3转录水平的改变。免疫印迹法检测survivin和caspase-3蛋白的表达情况。结果氧化苦参碱作用人膀胱癌细胞后,光镜及电镜下均见到典型的细胞凋亡形态,流式细胞术观察发现凋亡峰。同时观察到氧化苦参碱抑制survivin的转录和表达,对caspase-3有上调作用(P<0.05)。结论氧化苦参碱可能通过抑制survivin、上调caspase-3表达诱导膀胱癌T24细胞发生凋亡。Objective To observe the effect of plant alkaloid oxymatrine (OMT) on the prohieratlon and apoptosls ot bladder cancer cell line T24 and the possible molecular mechanism. Methods The effects of OMT (0. 625, 1.25, 2.5 mg/mL) on the viability and apoptosis of T24 cells were examined by optic, electron microscopy and flow cytometric analysis (FCM). The expression of survivin and caspase-3 genes was detected by RT-PCR, and the protein levels of survivin and caspase-3 were analyzed by Western blotting analysis. Results Typical apoptotic morphologies were observed under optic microscope and electron microscope, and the apoptosis peaks were observed by FCM after treatment with OMT. OMT significantly suppressed survivin expression and up-regulated caspase-3 expression (P〈0.05). Conclusion OMT can induce apoptosis of human bladder cancer T24 cell line by decreasing survivin expression and enhancing caspase-3 expression.
