摘 要:目的探讨槲皮素对宫颈癌侵袭和转移的作用机制及其与乙酰肝素酶(heparanase,HPA)的相关性。方法构建宫颈癌Hela细胞组、Hela+槲皮素组、HPA-SiRNA-Hela组、HPA-SiRNA-Hela+槲皮素组4组细胞模型,应用Transwell小室基质划痕损伤实验方法,测定不同时间点各组宫颈癌Hela细胞侵袭能力的变化。结果在6,12,24h时,Hela+槲皮素组及HPA-SiRNA-Hela组与Hela细胞组比较,宫颈癌Hela细胞迁移能力明显下降(P<0.01);HPA-SiRNA-Hela组与Hela+槲皮素组比较差异无统计学意义(P>0.05);HPA-SiRNA-Hela+槲皮素组与其他3组比较,宫颈癌Hela细胞迁移能力均下降(P均<0.01)。结论槲皮素对宫颈癌Hela细胞的侵袭能力有明显抑制作用,HPA与宫颈癌Hela细胞的侵袭过程相关,而槲皮素对宫颈癌Hela细胞的侵袭能力的抑制作用可能与HPA相关。Objective To investigate the mechanism of quercetin on the invasion and metastasis of cervical cancer and the relationship with heparanase (HPA). Methods Four cell model groups were constructed as cervical cancer Hela cell group (control group), quercetin treatment group, HPA gene silencing group and HPA gene silencing plus quercetin treatment group. Transwell small room method was used to observe the change of invasion ability of cervical cancer cell at different time points. Results In 6, 12, 24 hours, the migration of cervical cancer Hela cell decreased significantly in quercetin treatment group and HPA gene silencing group compared with control group (P〈0.01), and there was no significant difference between quercetin treatment group and HPA gene silencing group (P〉 0.05). The migration of cervical cancer Hela cell was lower in HPA gene silencing plus quercetin treatment group compared with the other groups (P〈 0.01). Conclusion Quercetin can significantly inhibit invasion ability of cervical cancer cell. Heparanase is correlated with the invasion process of cervical cancer cell, and the inhibition of quercetin in cervical cancer invasion ability may be correlated with HPA.
