摘  要:目的:研究双歧杆菌脂磷壁酸(LTA)对宫颈癌HeLa细胞体外生长的影响。方法 :HeLa细胞与不同浓度的LTA(10.0、20.0、40.0μg/L)共培养后EdU检测其增殖率;TUNEL法和RT-PCR分别检测LTA40.0μg/L共培养细胞的凋亡率及基因Bax/Bcl-2、Smac/IAP-1的表达变化。结果:10.0~40.0μg/L的LTA均能抑制Hela细胞增殖,作用与浓度正相关;LTA40.0μg/L共培养细胞48 h后凋亡率明显增加(P<0.05);Bax基因表达上调,Bcl-2基因表达下调(P<0.05),Smac/IAP-1无明显改变(P>0.05)。结论:LTA可能是通过调控Bax、Bcl-2的表达实现抑制HeLa细胞体外增殖并诱导其凋亡的。Objective To study the effects of lipoteichoic acid(LTA) on the growth of HeLa cells in vitro.Methods HeLa cells were co-cultured with various concentrations of LTA(10.0 μg / L,20.0 μg / L,40.0 μg / L).Then the growth of HeLa cells was detected by EdU staining.The apoptosis rate and the expressions of Bax / Bcl-2 and Smac / IAP-1 genes of HeLa cells co-cultured with 40.0 μg / L of LTA were detected by TUNEL staining and RTPCR respectively.Results The growth of HeLa cells could be inhibited by 10.0-40.0 μg / L of LTA,and 40.0 μg / L was the most significant.The apoptosis rate of HeLa cells co-cultured with 40.0 μg / L of LTA for 48h was increased significantly(P 0.05),and the expression of Bax gene was up-regulated,Bcl-2 gene was down-regulated(P 0.05),and Smac / IAP-1 had no significant change(P 0.05).Conclusion LTA can not only inhibit the growth of HeLa cells but also induce the apoptosis in votro through regulating the expressions of Bax and Bcl-2.
