摘 要:非遗传毒性致癌物在长期动物实验中常表现出致癌活性,因其作用机制不同于遗传毒性致癌物,在常规遗传毒性试验中表现出阴性。因此,有必要建立一种新的检测非遗传毒性致癌物的方法。两阶段细胞转化试验是在体外细胞转化试验的基础上加以改进的一种灵敏、有效的短期试验方法。该方法既可以检测遗传毒性致癌物,也可以检测非遗传毒性致癌物,具有快速、简便、经济的特点。近几年国内外多项研究均致力于适用该方法的新细胞系的建立,并采用了高通量筛选方法以及改进了结果判定的方法以提高结果的准确性和客观性。本文综述了近年来两阶段细胞转化试验方法的改进、发展及其应用前景。A considerable number of non-genotoxic chemicals have been shown to be carcinogenic in long- term animal experiments, however, these chemicals have negative results in routine genotoxic assays because of dif- ferent mechanisms of actions between genotoxic carcinogens and non-genotoxic carcinogens. Therefore, it is neces- sary to establish a new method to screen the non-genotoxic carcinogens. Two-stage cell transformation assay is a sensitive and effective short-term system, which is based on in vitro cell transformation assay. It can predict chemi- cal carcinogenicity including both genotoxicity and non-genotoxicity with the characteristics of simpleness, rapidness and cost-effectiveness. In recent years, a number of studies at home and abroad are all committed to the establish- ment of new cell lines for the experiment, application of high throughput screening and improvement of evaluation methods in order to improve the objectivity and accuracy of the results. This paper presents a review of the improve- ment and progress of two-stage cell transformation assay in recent years and its application prospect.
