摘 要:目的:使用TaqMan探针技术对IL-28B基因单核苷酸多态性进行基因分型,探讨IL-28B基因多态性与丙型肝炎病毒(HCV)易感性的相关性。方法:根据IL-28B基因rs8099917位点T〉G多态性,设计针对不同等位基因的TaqMan探针,借助实时PCR技术进行基因分型。采用病例-对照研究方法,对93例HCV患者和185例健康人群IL-28B基因rs8099917位点进行等位基因分型,比较两组人群基因型分布及频率差异。结果:TaqMan探针技术可以有效对rs8099917位点进行基因分型,研究人群中存在TT、TG、GG共3种基因型。T等位基因频率在HCV患者、健康人群中分别为90.3%和95.1%,差异有统计学意义(P〈0.05)。含G等位基因人群感染HCV的相对危险度OR值为2.095,95%CI为1.06~4.13。结论:IL-28B基因rs8099917位点与丙型肝炎易感性相关,G等位基因是HCV易感基因。Objective To explore the correlation between IL-28B gene Single Nucleotide polymorphisms and the susceptibility to Hepatitis C virus (HCV) by using TaqMan probe technology. Methods TaqMan probe was designed and real-time PCR was used to genotype IL-28B gene rs8099917 T 〉G pdymcrphism. A case-control study was used to detect rs8099917 T〉 G polymorphism, involving 93 HCV infected patients and 185 healthy controls. Then, the differences of genotype distribution and allele frequency between two groups were compared. Results TaqMan probe technology could be genotyped rs8099917 polymorphism effectively, and it existed TT,healthy controls respectively, and the difference was statistically significant (P〈 0.05). Populations with allele G had higher risk to get HCV infection with OR 2.095, and 95% CI 1.06-4.13. Conclusion IL-28B gene rs8099917 SNP was associated with the susceptibility to HCV, G allele was the susceptible allele to HCV.
