摘  要:目的 探索一种新的脑胶质瘤生长抑制因子Glob-N的分离鉴定方法。方法 人脑中性鞘糖脂经过加压柱层析,梯度甲醇/氯仿溶液洗脱。对所得到的产物行1H-NMR和13C-NMR测定。结果 含甲醇60%的甲醇/氯仿溶液的洗脱物为纯化Glob-N,层析展开后呈单一清晰条带,Rf值0.45。初步结构分析表明人脑Glob-N含N-乙酰半乳糖,共3个糖基,其神经酰胺部分共有33个碳原子。结论 柱层析分离中性鞘糖脂单一组分效果好,纯度高;人脑Glob结构中含3个糖基。Objective To develop a new method for isolating and identifying a glioma growth factor inhibitor, Glob-N, from normal human brain. Methods Neutral glycosylsphingolipid was separated from normal human brain by column chromatography with methanol/chloroform solution for gradient elution, and the resultant underwent 1H-NMR and 13C-NMR analyses for identifying the structure of Glob-N. Results Glob-N was obtained by elution with 60% (V/V) methanol in methanol/chloroform solution, with the Rf value of 0.45. The structural analysis showed that Glob-N from human brain was a globotriaosylceramide, containing GalNAc with 33 carbon atom in its ceramide moiety. Conclusion Individual N-GSL can be well separated from total N-GSL by column chromatography with high output. Glob-N from human brain maybe a kind of globotriaosylceramide.
