摘　　要：目的：研究苯并卟啉衍生物单环酸A（BPD）光动力杀伤人鼻咽低分化鳞癌CNE2细胞株的作用机理。方法：在体外培养的CNE2细胞株中加入不同浓度的BPD后，用690nm单色光照射仪照射，照光能量分别为1.2J/cm2、2.4J/cm2、4.8J/cm23组。MTT法检测细胞活性。并收集细胞进行电镜检查。结果：在同一光照能量下，不同浓度BPD对CNE2细胞的杀伤作用随BPD剂量的增大而增强，照光能量分别为1.2J/cm2、2.4J/cm2、4.8J/cm23组，其IC50值分别为1958.6ng/ml、506.38ng/ml和343.0ng/ml。电镜检查：BPD晚光组光照15min，细胞体积已比单纯光照射组缩小，整个细胞密度增加，胞质内所有细胞器呈轻度收缩状态；2h，一些细胞胞质出现明显自溶；8h后能见到2种形式的细胞死亡：坏死和细胞凋亡；24h，主要以坏死细胞为主。结论：在同一光照能量下，对CNE2细胞的杀伤作用随BPD剂量的增大而增强，光动力作用引起的肿瘤细胞死亡存在两种时相及方式，在同一条件下出现两种不同的时相及死亡方式，可能与细胞处在不同生长阶段，代谢程度不一，吸收BPD的含量不同有关。Objective： To investigate the killing mechanism of photodynamic effect using benzoporphyrin derivative (BPD) on CNE 2 cell line. Methods： The experiment of photodynamic killing effect on CNE 2 cell line was performed in vitro by using benzoporphyrin derivative and monochromatic light at wavelength around 690nm. The killing effect on CNE 2 cell line were evaluated by MTT assay and the cells were examined under electron microscope. Results： IC50 value of BPD with light energy of 1.2 J/cm2, 2.4 J/cm2 and 4.8 J/cm2 was 1958.6 ng/ml, 506.38 ng/ml and 343.0 ng/ml, respectively. Thirty minutes after irradiation, some tumor cells contracted and the density of cellular matrix was increased with shrinkage of the cellular organella. Two hours following irradiation, autolytic area of cytoplasm of CNE 2 tumor cells could be seen. Eight to sixteen hours after irradiation, both necrosis and apoptosis of tumor cells were presented. Twenty four hours after irradiation,necrosis of tumor cells became severe.Conclusions： At the same energy of irradiation, the killing effect on CNE 2 cells depend on the dosage of BPD. There are two patterns and time phase of tumor cells death after irradiation and BPD,which may be associated with the different growth stage, metabolism and BPD absorption of the cells.
