摘 要:目的:探讨FHIT基因与大肠癌的关系。方法:采用RTPCR及PCR产物直接测序法,对30例大肠癌组织及其配对正常组织、4个结肠癌细胞系的FHIT基因进行检测。结果:在8例(26.7%)大肠癌组织和2个(50%)结肠癌细胞系中检测到异常FHIT转录本,配对正常组织均无异常FHIT转录本;测序证实异常转录本缺失1~3个FHIT外显子。异常FHIT转录本与大肠癌患者的年龄、性别、血清CEA水平、肿瘤部位、大小、组织学类型、分化程度及病理分期均无关(P>0.100)。结论:在大肠癌发生中,异常FHIT转录本的表达是常见的事件;FHIT基因异常与大肠癌的发生发展有关。Objective: To evaluate the relationship between the FHIT gene and colorectal cancer. Methods: Matched normal and cancerous tissues from 30 patients with colorectal cancer and 4 human colorectal cancer cell lines were analysed by nested RT PCR and direct sequencing of the products. Results: A total of 8 of 30(26.7% ) primary colorectal cancerous tissues and 2 of 4 (50% ) colorectal cancer cell lines expressed aberrant FHIT transcripts, while no aberrant FHIT transcript was observed in all the matched normal tissues. Sequence analysis of the aberrant transcripts of 2 cancerous tissues were confirmed the absence of 1 to 3 exons of the FHIT gene. Clinicopathological analysis of patients showed that the aberrant FHIT transcripts were associated neither with age, sex and serum CEA levels of the patients, nor with sites, sizes, histological classification, differentiation grades and pathological stages of the tumors. Conclusions: Expression of aberrant FHIT transcripts is common in colorectal cancer and abnomalities of the FHIT gene transcripts are associated with colorectal carcinogenesis.
