摘 要:目的:为放射免疫显像在诊断食管癌淋巴结转移中的应用,提供基础研究支持。方法:①免疫组化(LSAB法)测定不同组织冷冻切片和抗人食管鳞癌单抗G9的反应;②125I标记G9,形成标记化合物,在荷食管癌裸鼠腹腔注射125I-G9后的连续3天内,测定各主要组织、器官的放射性。结果:①LSAB显示,食管癌原发灶和转移淋巴结呈阳性反应,正常食管和无转移淋巴结阴性。②125I-G9在裸鼠体内分布显示,肿瘤组织的放射性计数均明显高于其它器官/组织;各器官/组织的T/NT值在2~7之间;放射自显影的结果也显示,肿瘤部位有大量的放射性浓集。结论:①单抗G9能选择性定位于食管癌原发灶与转移淋巴结的癌细胞胞膜。②125I-G9在荷食管癌裸鼠有肿瘤组织导向作用,显像效果良好,提示放射免疫显像在食管癌转移淋巴结定位,有潜在的临床应用前景。Objective: Providing the experimental foundation for farther application of radioimmunoimaging of lymph node metastasis in esophageal carcinoma patient. Methods: Newly identified and prepared McAb G9 may be specific to cellular membrane antigen of esophageal carcinoma. Surgical samples frozen sections were detected by immunohistoche mistry (LSAB); 125I labeled G9 was prepared by Chloramine T method. The uptake radioactivitys of tumor and normal tissues in 15 tumor bearing nude mice were measured at different time (24, 48, and 72 h) after peritoneal injection of 125I-G9. Results: LSAB showing G9 reaction positively with the primary tumor and the metastatic lymph node, negatively with normal esophageal tissue and tumor free lymph node. The distributions of 125I-G9 in tumor during the three day showed by radioactivities were obviously superior to other organ/tissue (except blood) and the highest was at the 48 h(% ID/g 7.63). The T/NT values were 2~ 7. The autoradiography indicated that radioactivity markedly concentrated in tumor tissue. The concentration in the edge is more obviously than that in other part of the tumor. Conclusion: McAb G9 is specific to cellular membrane antigen of esophageal carcinoma, either in the primary lesion and metastases of lymph node. 125I-G9 has a considerable targeting activity and can well locate in esophageal carcinoma tissue in tumor bearing nude mice so that could be potentially applied in radioimmunoimaging for detecting lymph node metastasis in esophageal carcinoma patient.
