摘  要:目的探讨Rock-1的表达对宫颈癌侵袭能力的影响。方法采用RT-PCR和Westernblot方法检测正常宫颈组织、宫颈癌细胞株Hela及其以Rho激酶特异性抑制剂Y-27632处理后的Rock-1和磷酸化cofilin表达变化。并用Transwell小室体外侵袭方法,检测Y-27632处理前、后Hela细胞体外侵袭能力的变化。结果宫颈癌细胞株Hela细胞中,Rock-1和磷酸化cofilin的表达量均高于正常宫颈组织(P<0.05)。不同浓度的Y-27632阻断Rock-1作用后,Hela细胞中磷酸化cofilin的表达量及体外侵袭能力降低(P<0.05)。结论 Rock-1在宫颈癌细胞中的表达高于正常宫颈组织;抑制Rock-1的表达能降低宫颈癌细胞株Hela细胞的体外侵袭能力。Objective To investigate the effect of Rock-1 expression on the invasion ability of cervical carcinoma cell in vitro. Methods The expression of Rock-1 and phosphorylated cofilin in normal cervical tissue, cervical carcinoma Y-27362 Hela cells before and after treated with Rho kinase were measured by RT-PCR and Western blot. Transwell assay was used to detect the changes of in vitro invasion capacity of Y-27362 Hela cells before and after treated with Rho kinase in vitro. Results The expressions of Rock-1 and phosphorylated cofilin in cervical carcinoma Hela cells were higher than those in normal cervical tissues(P〈0. 05). The expression level of phosphorylated cofilin and in vitro invasive ability of Hela cells were reduced after treated with Rho kinase(P〈0. 05). Conclusian Rock-1 expression in cervical carcinoma cells is higher than that in normal cervical tissues. In vitro invasive capacity of Hela cells is reduced by inhibiting Rock-1 expression in cervical carcinoma Hela cells.
