摘  要:目的:观察创伤后应激障碍(PTSD)模型大鼠海马神经元自噬的改变,探讨PTSD致海马体积异常的可能机制。方法:成年健康雄性SD大鼠20只,随机被分为对照组和模型组。采用改良的单一连续应激后给予不可逃避足底电击方法制备PTSD大鼠模型;采用透射电镜技术观察海马神经元超微结构,Western blot方法检测海马微管相关蛋白轻链3(microtube-associated protein 1 light chain 3,LC-3)和Beclin-1的表达水平。结果:模型组海马神经元自噬体增多;海马组织LC3-Ⅱ/LC3-Ⅰ比值和Beclin-1表达水平高于对照组(P<0.05)。结论:PTSD模型大鼠海马神经元存在明显的细胞自噬,可能与海马体积异常变化有关。Objective: To observe the change of autophagy in hippocampal neurons of rat with post-straumatic stress disorder (PTSD), and to investigate the possible mechanism of hippocampal volume alteration in PTSD. Methods: Twenty adult male SD rats were randomly divided into control group and PTSD model group. The PTSD animal model was established by using the modified single-prolonged stress followed by a single inescapable electric foot shock( SPS & S). The uhrastructure of neurons in hiappoeampus was observed under electron microscope. The expression of LC-3 and Beclin-1 protein in hippocampus was detected by Western blot. Results: Autophagosomes increased in hippocampal neurons of PTSD model rats. The ratio of LC3-Ⅱ to LC3-Ⅰ and the expression of Beclin-1 protein were higher in model rats than in control rats ( P 〈 0.05 ). Conclusion : Atuophagy was enhanced in hippocampal neurons in rats with PTSD, which might be related to the abnormal change of hippocampal volume in PTSD.
