龟象膏大鼠皮肤长期毒性实验研究

摘　　要：目的观察和测定龟象膏长期连续经皮肤给药对SD大鼠的长期毒性作用。方法将154只SD大鼠随机分为7组,即空白对照组,完整皮肤高、中、低剂量组及破损皮肤高、中、低剂量组,每组各22只,雌雄各半。完整皮肤、破损皮肤高、中、低剂量组分别在剃毛区涂龟象膏20、10、5 g/(只.d),空白对照组涂乳膏赋形剂凡士林10 g/(只.d)。涂药后用胶布固定,每日1次,连续90 d(每周停药1 d)。观察大鼠一般性状,计算给药前后体质量增减率。心脏采血,检测血常规及血液生化指标,随后做系统尸检及病理检查。结果完整皮肤、破损皮肤高、中、低剂量组大鼠外观、行为体征、活动、精神状态均无明显异常;饮食、粪便及尿液正常,体质量正常增加,体质量增减率之间比较差异均无统计学意义(P>0.05);无大鼠死亡情况发生,亦无中毒反应;主要血液学指标、生化指标、主要脏器系数与空白对照组比较差异均无统计学意义(P>0.05);解剖完整皮肤高剂量组、破损皮肤高剂量组及空白对照组大鼠主要脏器未见病理性改变。结论龟象膏长期连续经皮肤给药对实验大鼠无毒性反应,可以在临床安全使用。Objective To observe the long-term toxicity for applying Guixiang cream on rat skin Methods 154 SD rats were randomly divided into seven groups,blank control group,high-,middle-and low-dose groups of intact skin,and high-,middle-and low-dose groups of damaged skin.There were 22 SD in each group,male and female half and half.20 g/d of Guixiang cream were applied at high-dose group of intact skin and damaged skin.10 g/d of Guixiang cream were applied at middle-dose group of intact skin and damaged skin.And 5 g/d of Guixiang cream were applied at low-dose group of intact skin and damaged skin.The site of administration was fixed with adhesive tape,once a day,continuous 90 d（withdrawal one day a week）.The general character of the rat was observed,the rate of change of body mass was calculated before and after administration.Cardiac blood was collected,blood biochemical parameters were measured,followed by systematic autopsy and pathological examination.Results Rats＇ characters（appearance,behavior,signs,activities and mental state） had no change,the results of all detections including diet,feces and urine was normal.There was no significant difference between test groups and control group on main blood biochemical parameters,organ coefficient.Rats＇ major organs had no change in anatomy and pathology.Conclusion No long-term toxic reaction can be found in the test of rats applied Guixiang cream on their skins for a long term.It proves that it is safe using the normal dose of Guixiang cream on the skin in clinic.
