摘 要:目的探讨回药爱康方含药血清对人肺腺癌A549细胞生长的抑制作用及对细胞形态的影响。方法采用血清药理学方法,将SD大鼠分为对照组、爱康方低、中、高剂量组、环磷酰胺化疗组、回药爱康方低、中、高剂量分别联合环磷酰胺化疗组,制备含药血清;培养人肺腺癌A549细胞,将细胞分为实验组和对照组,实验组分别给予含5%、10%、20%浓度的相应各组含药血清,对照组给予等浓度的对照组大鼠血清,培养24、48、72h后观察细胞形态变化,采用MTT法测定各组含药血清对A549细胞的生长抑制率。结果光镜下观察到,与对照组相比,各剂量回药爱康方含药血清可使A549细胞有不同程度的胞膜回缩、胞核固缩、细胞间距增大和染色质边缘化现象,其中以爱康方高剂量联合环磷酰胺组作用72h最明显。MTT法检测细胞生长抑制率发现,各实验组各浓度抑制率与相应对照组比较,实验组72h抑制率均高于对照组(P<0.05);单纯环磷酰胺72h抑制率均高于爱康方低、中、高剂量(P<0.05);爱康方高剂量联合环磷酰胺72h抑制率高于爱康方低剂量联合环磷酰胺和单纯环磷酰胺(P<0.05)。结论回药爱康方可有效抑制A549细胞生长增殖,并增强环磷酰胺对A549细胞的抑制作用。Objective To observe the effect of medicated serum with Hui Aikang Fang on growth and prolifer-ation of human lung adenocarcinoma A549 cells. Methods Using the serum pharmacology, SD rats were di-vided into low, medium, high dose Aikang Fang group, Cyclophosphamide(CTX) group, low, medium, high dose Aikang Fang combined with CTX group and normal saline group. The medicated serum with different concentrations of Aikang Fang were prepared. The ceils were divided into experimental and control groups : the experimental group were treated with 5%, 10%, 20% concentration corresponding group medicated serum while control group was given the same concentration of control serum. The morphological changes of the ceils were observed at 24h, 48h, 72h in each group. The proliferation of A549 was measured by methyl thiazolyl tetrazolium (MTT) method. Results Observed under the microscope and compared with the control group, the distance between the cells were increased, nuclear condensation, the chromatin concentrated to edge in Aikang Fang groups. By MTT method, compared with control group, all medicated serum groups' inhibition rates were significantly higher at 72 hours ( P 〈 0.05 ). The inhibition rate of CTX group were significantly higher than that of low, middle, high dose of Aikang Fang group at 72 hours (P 〈 0.05 ). The inhibition rate of high dose Aikang Fang combined with CTX were significantly higher than that of low dose Aikang Fang com- bined with CTX and CTX at 72 hours ( P 〈 0.05 ). Conclusion Hui Aikang Fang can inhibit proliferation and strengthen the curatiw effect of CTX on A549 cells.
