摘 要:目的检测PI3K、PTEN和Akt蛋白在肾透明细胞癌组织中的表达,分析三者之间的相关性及其表达与肾透明细胞癌临床病理特征的关系。方法采用免疫组织化学SP法检测60例肾透明细胞癌组织和癌旁组织中PI3K、PTEN和Akt的蛋白表达情况,分析三种蛋白的表达与肾细胞癌临床病理特征之间的相关性。结果①肾透明细胞癌组织中PI3K和Akt蛋白的阳性表达率分别为81.67%(49/60)和78.33%(47/60),明显高于癌旁组织中的10%(6/60)和15%(9/60)(P<0.05);PTEN蛋白在肾透明细胞癌组织中的阳性表达率为38.33%(23/60),明显低于癌旁组织中的88.33%(53/60)(P<0.05)。②在60例肾透明细胞癌组织中不同年龄、不同大小肿瘤的PI3K、AKT、PTEN蛋白表达阳性率差异无统计学意义(P>0.05)。而在病理分级、临床分期方面,三种蛋白表达阳性率差异有统计学意义(P<0.05);③PTEN与PI3K、Akt蛋白表达之间均呈显著负相关(r=-0.338和r=-0.325),PI3K与Akt蛋白表达呈显著正相关(r=0.391)。结论 PI3K,Akt蛋白在肾透明细胞癌组织中高表达,而PTEN则存在高比例丢失现象。Objective To detect PI3K, PTEN and Akt protein expression in renal clear cell carcinoma tissues and to analyze the correlation between expression and and its clinical pathological features. Methods The ex-pression of PI3 K protein, AKT protein and PTEN protein in renal clear cell carcinoma and normal renal tissues were detected by immunohistochemical method. Results ①The positive expression rate of PI3K and AKT protein in renal clear cell carcinoma was 81.67% (49/60) and 78.33% (47/60) respectively and they were significantly higher than those in the tissues adjacent to carcinoma( 10% and 15% , P 〈0.05). PTEN protein positive expression rate in renal clear cell carcinoma tissue was 38.33% (23/60) and it was significantly low-er than that in the tissues adjacent to carcinoma (88.33%, P 〈 0.05). ②The protein expression of PI3K, AKT and PTEN did showed significant difference in different age and size of tumor (P 〉 0.05 ) while signifi- cant difference of expression of PI3K, AKT and PTEN was found in different pathological grades and clinical stages (P 〈0.05). ③PTEN, PI3K and Akt protein expression showed significantly negative correlation(r = -0. 338 and r = -0. 325 ) while PI3K and Akt protein expression was significantly positive correlation (r = 0. 391 ). Conclusion PI3K and Akt protein show high expression in renal clear cell carcinoma tissues. PTEN shows loss high proportion in renal clear cell carcinoma tissues and it plays an important role in occur-rence and development of renal clear cell carcinoma.
