过继转移日本血吸虫感染鼠CD8α^-CD11c^+型DC亚群对OVA诱导的CCL-11和IL-13Rα_2的影响

摘  要:目的:通过过继转移日本血吸虫感染鼠树突状细胞(DC)亚群,探讨DC亚群对卵清蛋白(OVA)诱导的肺组织CCL-11和IL-13Rα2表达及嗜酸性粒细胞浸润的影响。方法:用CD8α和CD11c磁珠分离纯化日本血吸虫感染鼠DC亚群,分别获得CD8α-CD11c+DC和CD8α+CD11c+DC。将24只BALB/c小鼠随机分为4组:正常对照组(Normal组),单纯诱发哮喘组(OVA组),过继转移日本血吸虫感染鼠CD8α+CD11c+DC并诱发哮喘组(SJCD8α+DC/OVA组)和过继转移日本血吸虫感染鼠CD8α-CD11c+DC并诱发哮喘组(SJCD8α-DC/OVA组)。SJCD8α+DC/OVA和SJCD8α-DC/OVA两组小鼠分别经尾静脉过继转移相应的DC亚群,1 h后除正常组外均开始诱发哮喘。4周后处死小鼠,取小鼠肺组织制成切片,进行HE染色和免疫组化染色,观察炎性变化,检测CCL-11和IL-13Rα2在肺组织中的表达。结果:肺组织切片HE染色显示Normal组小鼠无炎症,SJCD8α-DC/OVA组小鼠炎症较轻,SJCD8α+DC/OVA和OVA组炎症明显且有大量炎性细胞浸润。免疫组化结果显示,Normal、OVA、SJCD8α+DC/OVA和SJCD8α-DC/OVA 4组小鼠肺组织CCL-11阳性区域平均光密度值分别为0.152 3±0.010 9、1.496 7±0.225 6、1.343 5±0.094 9和0.863 4±0.211 4,CCL-11阳性区域面积占血管、气管总面积的百分比分别为0.000 4±0.000 3、0.257 2±0.085 9、0.210 3±0.023 1和0.039 2±0.005 4;IL-13Rα2阳性区域平均光密度值分别为0.152 7±0.015 3、0.334 5±0.003 7、0.283 2±0.035 0和0.175 5±0.020 1,IL-13Rα2阳性区域占血管、气管总面积的百分比分别为0.037 0±0.034 1、0.297 0±0.006 2、0.287 0±0.031 0和0.117 1±0.005 0。差异均有统计学意义(P均<0.05)。结论:日本血吸虫感染鼠CD8α-CD11c+DC亚群可以抑制OVA诱导的CCL-11和IL-13Rα2表达,从而对哮喘起到抑制作用。Objective: To study the effect of the dendritic cells (DCs) subsets infection on CCL-11 and IL-13Rα2 expression and eosinophils infiltration in OVA-induced lung tissue cells by adoptive transfer of DC subsets from mice infected with Schistosomajapon- icum. Methods: DC subsets from mice infected with S. japonicun were respectively isolated and purified using CD8α and CD1 lc mi- crobeads. Twenty four BALB/c mice were randomly divided into four groups. Group normal served as the control. Group OVA was the asthma group. Each mouse in group SJCD8α+DC/OVA was injected with CD8α+DC from mice infected S.japonicun, while each mouse in group SJCD8α-DC/OVA was injected with CD8α-DC. One hour after adoptive transfer, All but group normal were sensitized and chal- lenged with ovalbumin to induce asthma. Mice were sacrificed after four weeks and their lungs were removed for HE staining and im- munohistochemistry staining, inflammatory changes were observed, and levels of CCL-11 and IL-13Rα2 expression were detected in lung tissue. Results:HE staining indicated that group Normal had no inflammation and SJCDSα-DC/OVA had slight inflammation. The groups SJCD8α +DC/OVA and OVA had serious inflammation and larger number of infiltrating inflammatory cells. In group Nromal, OVA, SJCDSα+DC/OVA and SJCD8α-DC/OVA lung tissue immuohistochemical staining showed that the average opitical density(AOD) of CCL- 11 were 0.152 3±0.010 9,1.496 7±0.225 6, 1.343 5±0.094 9 and 0.863 4±0.211 4, the positive area of CCL-11 in blood vessels and tra- chea's percentage of the total area were 0.000 4±0.000 3, 0.257 2±0.085 9, 0.210 3±0.023 1 and 0.039 2±0.005 4; the AOD of IL-13Rα2 were 0.152 7±0.015 3, 0.334 5±0.003 7, 0.283 2±0.035 0 and 0.175 5±0.0201, the percentage of the IL-13Rα2's area of the total area were 0.037 0±0.034 1, 0.297 0±0.006 2, 0.287 0±0.031 0 and 0.117 1±0.005 0.
