摘  要:目的观察IL-13对急性肾缺血再灌注损伤(RIRI)大鼠IL-6表达的影响。方法 Wistar雄性大鼠57只,随机分为5组:假手术组、缺血再灌注组、治疗对照组、治疗1组和治疗2组。阻断大鼠双侧肾脏血流45 min再灌注24 h建立急性RIRI模型;于阻断血流后分别从双侧肾动脉开口注射入鼠重组IL-13(rmIL-13)治疗1组0.5 g/50 g,治疗2组1.5g/50 g;治疗对照组以生理盐水代替。检测各组大鼠血清IL-6水平和肾脏表达,以及肾功能和肾脏病理。结果与治疗对照组比较,治疗2组肾脏IL-6基因和蛋白表达明显减少(P<0.01),IL-6血清水平也明显下降(P<0.01);治疗2组肾功能障碍及BUN、Cr水平显著改善(P均<0.01),肾组织病理变化明显减轻,肾小管损害评分明显减少(P<0.01)。血清IL-6蛋白水平与BUN、Cr呈正相关(r=0.628,0.745,P<0.01),肾组织IL-6 mRNA与BUN、Cr呈正相关(r=0.745,0.792,P<0.01)。结论 IL-13能有效地抑制急性RIRI大鼠IL-6的表达,并保护肾功能。Objective It is to observe the influence of IL - 13 on expression of IL - 6 in rat with acute renal ischemia reperfusion injury (RIRI). Methods 57 Wistar rats of male were randomly divided into 5 groups: pseudo surgery group ( Group sham) , ischemia-reperfusion groups ( Group I/R) , treatment control group ( Group C) , treatment 1 group ( Group T1 ) and treatment 2 group ( Group T2). The RIRI model was made by blocking bilateral renal blood flow 45 min and perfusion 24 h in rats. The animals were injected with recombinant interleukin 13 ( rmIL - 13) of 0.5 g/50 g in Group T1 and 1.5 g/50g in Group T2 after blocking blood flow from bilateral renal artery, and was injected with saline solution in Group C. The levels of IL - 6 in serum and kidney in rat were detected, and renal function and renal pathology were tested in all groups. Resuits Compared with Group C, in Group T2, the gene and protein expression of IL - 6 in renal were markedly reduced ( P 〈 0.01 ), and the levels in serum was significant decreased (P 〈0.01 ); the kidney dysfunction, levels of BUN and Cr were markedly improved ( all P 〈 0.01 ) , and renal pathological changes were remarkably decrease, renal tubular damage and score were significantly reduced ( P 〈 0.01 ). The levels of IL - 6 protein in serum was positively correlated with BUN and Cr ( r = 0. 628, P 〈 0. 01 ; r = 0. 745, P 〈 0.01 ) , and IL - 6 mRNA in renal tissues was positively correlated with BUN and Cr ( r = 0. 745, P 〈0.01 ; r =0. 792, P 〈0.01 ). Conclusion IL - 13 can effectually inhibit the IL -6 expression in acute RIRI rat, and protect the renal function.
