摘  要:目的:对艾烟中可吸入颗粒物冷凝液(MSC)进行鼠伤寒沙门氏菌回复突变试验,评价艾烟的潜在致突变性。方法:采用平板掺入法观察艾烟引发试验选用菌株TA97、TA98、TA100、TA102的回变菌落数。试验设0、62.5、125、250、500、1 000μg/皿,阳性对照组,分别做代谢系统活化及非活化的对照,每组设3个平行。在背景生长良好的条件下,计数受试物组的菌落回变数,回变菌落数等于或大于2乘以未处理对照数并有剂量反应关系或至少某一测试点有可重复的并有统计学意义的阳性反应,即可认为该受试物诱变试验阳性。结果:MSC在活化下TA98诱变阳性,而对TA97、TA100、TA102活化以及非活化、TA98非活化均为阴性,此结果可以重复并且差异有统计学意义。结论:MSC使细菌生长增多,并且有剂量反应关系,根据本实验结果MSC具有潜在的致基因突变性。Objective: Screening of moxa smoke condensation (MSC) for mutagenicity by using the Ames test to detect the producing genetic damage that leads to gene mutations. Methods: Using the plate incorporation assay, MSC were assayed for mutagenicity employing several histidine dependent Salmonella strains (TA97, TA98, TA100 and TA102) both with and without metabolic activation using a liver fraction (S-9). Treatment group, control group, 62.5, 125, 250, 500, 1 000μg/dish, positive control, control group+S9, 62.5+S9, 125+S9, 250+S9, 500+S9, 1 000μg/dish+S9, positive control+S9, and each group set up three plate. When a mutagen was added to the plate, the number of revertant colonies per plate increased, equal to or 2 times more than the number of spontaneously induced revertant colonies per plate, usually in a dose-related manner. Results: Without S-9, MSC showed that mutagenic activity was not enhanced; with S-9 MSC, it was found to be mutagenic for strain TA98 rather than TA97, TA100, TA102. Conclusion: MSC increased Salmonella growth, and there are dose-response relationship. The Ames test screened that MSC was a potential mutagenic.
