S100蛋白表达与鼻咽癌临床关系的研究

摘　　要：目的探讨S100蛋白家族在鼻咽癌组织中的表达及临床意义。方法应用免疫组织化学方法研究S100蛋白家族成员(S100A2、A4、A9、A11)和肿瘤侵袭相关蛋白基质金属蛋白酶-2(MMP-2)在58例低分化鼻咽癌患者的癌组织及16例正常鼻咽组织中的表达。结果 58例癌组织中S100A4和S100A9阳性的表达率分别是78%和81%,明显高于其在正常鼻咽组织中的表达(P<0.01),而S100A2在鼻咽癌组织中的表达明显低于正常鼻咽组织(P<0.01),S100A11则在鼻咽癌组织中的表达同正常鼻咽组织相比无明显差异(P>0.05)。在S100A4+/S100A9+/S100A2-的癌组织中MMP-2的表达明显高于其他组织(P<0.05)。结论 S100A4和S100A9蛋白的高表达与鼻咽癌的发生发展呈正相关,而S100A2蛋白则能抑制鼻咽癌的发生,S100A11蛋白与鼻咽癌的发生无相关性。S100A4、S100A9和S100A2可能是通过MMP-2的上调促进了鼻咽癌细胞的恶性侵袭能力。Objective To detect the experssion of S100 protein family in nasopharyngeal carcinoma and analysis the correlation. Methods Immunohistochemical analysis used in 58 cases of NPC and 16 cases of normal nasopharyngeal mucosa （NNP） ,the data analyzed statistically with SPSS. Results The expression level of S100A4, A9 proteins were significantly up- regulated in the stroma of NPC compared to that in NNP （P〈0.01）. On the contrary, S100A2 was silenced. The expression level of S100A11 in NPC was the same of the NNP. Conclusion The expression of S100A2, A4, A9 protein closely relate to the occurrence of nasopharyngeal carcinoma, which they promote the malignant invasion ability of the nasopharyngeal carcinoma cells maybe through the raising of MMP-2.
