摘 要:目的探讨内质网应激(ERS)的激活剂衣霉素(Tm)能否激活心肌细胞的氯通道,并观察其电生理学特性。方法原代培养新生SD大鼠心肌细胞,用等渗外液灌流作为对照,然后改变灌流液为低渗或者含有衣霉素的等渗外液,同时通过膜片钳全细胞模式记录电流。结果 (1)在等渗外液下,仅记录到低幅度的基础电流。与等渗外液相比,低渗外液灌流细胞时电流的密度在+40、+60、+80及+100mv时明显增加(P<0.05),并且电生理学特性类似于容积敏感氯通道(VSOR)的电流,包括:外向整流、正电压依赖性失活,以及对氯离子通道阻断剂4,4’-异二硫氮氏-2,2’-二磺酸(DIDS)敏感。(2)与等渗外液对比,含有衣霉素(2.5μg/ml)的等渗外液灌流同样可以记录到电流密度的显著增加,电生理学特性类似于低渗外液诱导的VSOR氯通道电流。结论衣霉素可以诱发乳鼠心肌细胞的氯通道电流,并且符合VSOR的电生理学特性。Objective To explore whether the tunicamycin can activate the chloride channel and observe its electrophysiological properties in cardimyocyte.Methods perfusion of Cultured neonatal mouse cardiomyocytes with isotonic extracellular fluid as the control,then change the extracellular fluid to the hypotonic or the isotonic solution containing Tm(2.5μg/ml).The whole-cell patch clamp technique was performed to analyze the currents.Results We found that:(1)in compared with the isotonic solution,under the perfusion of the hypotonic,the current density significantly increased at +40,+60,+80 and +100 mv(P 0.05),and the properties of currents were similar to the volume-sensitive chloride channel,including:outwardly rectifying,inactivation in high large positive potential and sensitivity to the 4,4’-diidothiocya-natostilbene-2,2’-disulfonicacid(DIDS).(2) compared with the isotonic,the tunicamycin-induced currents were Similar to the properties of VSOR which recorded under the hypotonic.Conclusions These findings suggest that the tunicamycin can induce chloride currents in the cardiomyocyte which are consistent with the properties of VSOR chloride channel.
