肿瘤坏死因子-α对乳鼠心脏成纤维细胞增殖及syndecan-4蛋白表达的影响

摘　　要：目的观察肿瘤坏死因子-α(TNF-α)对乳鼠心脏成纤维细胞增殖及syndecan-4蛋白表达的影响。方法采用胰酶消化法体外培养SD乳鼠的心脏成纤维细胞,分别用质量浓度为5、10、20、30 ng/mL TNF-α作用24 h,并设立空白对照组进行比较。采用MTS/PMS法检测TNF-α对心脏成纤维细胞增殖的影响;Western blot法检测TNF-α对心脏成纤维细胞syndecan-4蛋白表达的影响。结果与对照组比较,5、10 ng/mL TNF-α对心脏成纤维细胞增殖及细胞syndecan-4蛋白表达无明显影响(P>0.05);而20、30ng/mL TNF-α均能促进心脏成纤维细胞增殖及细胞syndecan-4蛋白的表达(P<0.05),但不存在剂量依赖性。结论一定剂量的TNF-α可以促进心脏成纤维细胞的增殖及细胞syndecan-4蛋白的表达。Objective To investigate the effect of tumor necrosis factor-α（TNF-α） on the cell proliferation and syndecan-4 expression in neonatal rat cardiac fibroblasts.Methods Cardiac fibroblasts of neonatal SD rats were isolated by enzymatic dissociation and stimulated by 5,10,20 or 30 ng/mL TNF-α for 24 h.The ratio of proliferation of neonatal rat cardiac fibroblasts was determined by MTS/PMS assay and the expression of syndecan-4 protein was detected by western blotting.Results Compared with the control group,TNF-α at the concentration of 20 or 30 ng/mL,but not 5 or 10 ng/mL,significantly stimulated the proliferation of cardiac fibroblasts（P0.01）.Compared with the control group,TNF-α at the concentration of 20 or 30 ng/mL,but not 5 or 10 ng/mL,obviously enhanced the expression of syndecan-4 protein in cardiac fibroblasts（P0.05）.Conclusion TNF-α can stimulate the proliferation and the expression of syndecan-4 protein in neonatal rat cardiac fibroblasts.
