胃癌细胞腹腔种植模型的胃小肠Cajal间质细胞表达的分析

摘　　要：目的观察比较胃小肠Cajal间质细胞的表达。方法选取健康雌性裸鼠18只,随机分成正常对照组、胃癌细胞培养液组(培养液组)和胃癌细胞腹腔种植组(模型组),每组6只。分别于术后的3周,5周采集标本。采用免疫荧光法检测各组裸鼠胃、小肠壁层中c-Kit的表达并用软件测量其灰度值。结果①模型组、胃癌细胞培养液组胃壁c-Kit与正常对照组相比蛋白表达水平降低(P<0.01);②模型组、胃癌细胞培养液组小肠壁c-Kit与正常对照组相比蛋白表达水平降低(P<0.01);③小肠壁的Ca-jal间质细胞数量的减少明显高于胃壁的Cajal间质细胞数量的减少(P<0.05)。结论胃癌晚期的胃肠蠕动减慢与小肠壁中Cajal间质细胞的减少有直接的关系。Objective To observe and compare the expression of Cajal interstitial cell in stom- ach and small intestine. Methods Eighteen healthily female nude mice were randomly divided into normal control group, gastric cancer cell culture medium grbup （culture medium group） and gastric cancer cell peritoneal group （model group） （n = 6）. Specimens were collected af- ter 3 w, 5 w, respectively. The expression of c-Kit in the stomach and small intestine was de- tected by immunofluorescenee, gray value was measured by measurement software. Results ① Model group, gastric cancer cell culture medium group compared with the normal control group, the stomach wall protein expression levels of c-Kit were reduced（P 〈0. 01 ）. ② Model group, gastric cancer cell culture medium group compared with the normal control group, the small intestine c-Kit protein expression levels were decreased （ P 〈 0. 01 ）. ③ The amount of the decrease in interstitial cells of Cajal of the small intestine was greater than that in interstitial cells of Cajal of the stomach wall（ P 〈 0. 05 ）. Conclusion Advanced gastric cancer peristalsis slow down the direct relationship with the decrease in the small intestine of the interstitial cells of Cajal.
