摘  要:目的:检测瞬时受体电位M7通道(transient receptor potential melastatin 7,TRPM7)在人牙髓干细胞(hDPSCs)及牙髓组织的表达。方法:酶消化法分离培养hDPSCs,体外鉴定其表型特点及生物学特性;采用RT-PCR检测TRPM6、TRPM7mRNA在hDPSCs中水平的表达,Western blot检测TRPM7蛋白在hDPSCs中的表达;细胞免疫荧光检测TRPM7在hDPSCs的表达位置,免疫组化法检测TRPM7在牙髓组织中的表达。结果:分离培养的hDPSCs来源于间充质组织并且具有克隆形成和多向分化能力。hDPSCs表达TRPM6和TRPM7mRNA,表达TRPM7蛋白;TRPM7主要表达于hDPSCs的胞膜与胞浆;TRPM7在牙髓组织广泛表达。结论:hDPSCs和牙髓组织均表达TRPM7。AIM : To investigate the expression pattern of transient receptor potential melastatin 7 ( TRPM7 ) in human dental pulp stem cells (hDPSCs) and pulp tissues. METHODS: hDPSCs were obtained by collagenase di- gestion and in vitro culture. The hDPSCs were verified by flow cytometry and muhi-directional differentiation assay. RT-PCR was used to detect TRPM6 and TRPM7 mRNA level and Western blot was used to detect TRPM7 protein ex- pression in hDPSCs. TRPM7 protein in hDPSCs was examined by immunofluorescence and in dental pulp tissue by im- munohistochemistry. RESULTS: The cultured hDPSCs were proved to be originated from mesenchymal tissue, posses- sing colony-forming ability and multilineage differentiation potential. TRPM6 and TRPM7 mRNA and TRPM7 protein were expressed in hDPSCs, TRPM7 protein was localized in the cell membrane and cytoplasm. TRPM7 protein was widely distributed in pulp tissues. CONCLUSION: TRPM7 is expressed in hDPSCs and human dental pulp tissues.
