摘 要:目的:探索应用激光显微切割(Laser microdissection,LMD)技术获取小鼠牙胚特定部位组织的可行性。方法:选取胚胎期和出生后等多个时期CD1小鼠,取材后制作冰冻切片。经甲酚紫染色后再以莱卡LMD6000激光显微切割系统在高倍镜下分离获取下颌第一磨牙牙囊和牙乳头组织;下颌切牙根端上皮组织及其外围间充质组织。结果:应用LMD技术可实现对小鼠下颌第一磨牙及切牙胚胎期、出生后多个时期牙胚特定组织的精确分离。结论:LMD技术可用于分离获取相对均一的小鼠切牙、磨牙牙胚特异组织。AIM: To investigate the possibility of precise isolation of various tissues from murine tooth germ by laser mierodissection (LIVID) system. METHODS: Frozen sections of the dental follicle and dental papilla tissues of the first lower molar, and the apical bud epithelial tissues and mesenchyma tissues around the lower incisor of CD1 mice in embryonic and postnatal stages were prepared and dyed by cresyl violet then precisely isolated by LMD 6000 system, Laiea. RESULTS: Various epithelial and mesenchyma tissues of the mouse tooth germ in several embryonic and postnatal stages were successfully isolated by LMD system. CONCLUSION: LMD system can be used to acquire pure target tissues from tooth germ precisely.
