摘 要:目的采用高效液相色谱-蒸发光散射检测法(HPLC-ELSD法)建立同时测定广州水果中果糖、葡萄糖和蔗糖的方法。方法高效液相色谱条件为KromasilNH2(4.6mm×250mm,5μm)色谱柱,25℃柱温,流动相为乙腈-水(80/20,v/v)溶液,流速为1.5ml/min;蒸发光散射检测器(ELSD)漂移管温度为85℃,空气流速为2.5ml/min。结果果糖、葡萄糖和蔗糖的线性范围分别为0.6~10.0g/L、1.0~20.0g/L、0.6~20.0g/L;样品中3种糖的加标回收率分别为95.36%、98.44%、97.76%。结论首次建立了同时测定水果中果糖、葡萄糖、蔗糖的HPLC-ELSD法,为建立广州水果糖含量数据库提供了一种准确、灵敏、快速的分析方法。Objective To establish a high-performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD) method for quantifying the simultaneous quantitative analysis of fructose, glucose, sucrose in Guangzhou fruits. Methods The analysis was carried out using a Kromasil NH2 column (4.6 mm×250 mm, 5 μm) with a mobile phase composed of acetonitrile:water (80/20,v/v) at a flow rate of 1.5 ml/min. The column temperature was 25℃. The ELSD drift tube temperature was 85℃ and the flow rate of the dry air was 2.5 L/min. Results The linear range of detection of fructose,glucose and sucrose was 0.6-10.0 g/L, 1.0-20.0 g/L,and 0.6-20.0 g/L,respectively.The recoveries of these three sugars were 95.36%,98.44% and 97.76%. Conclusion This is the first study to establish a method for determining fructose, glucose and sucrose in Guangzhou fruits, and the method can be used in the building of sugar content database for Guangzhou fruits.
