摘 要:[目的]筛选高产酸性木聚糖酶细菌,并研究其适宜发酵条件。[方法]采用透明圈法筛选产木聚糖酶活力较高的细菌,并利用单因素试验与正交试验相结合的方法对其发酵条件进行优化。[结果]试验筛选到一株高产酸性木聚糖酶细菌B26,其最佳产酶培养基为:麸皮2%、秸秆粉2%、KNO30.8%、TW-80 0.6%,此时该菌所产木聚糖酶活性达1 051.25 U。[结论]该菌株以农业废弃物秸秆和麸皮为原料,产生活性较高的酸性木聚糖酶,对于农副产品的精深加工及利用开发有重要意义。[ Objective] The paper was to screen bacteria with high-producing acidic xylanase activity, and to study its optimal fermentationconditions. [ Method] The bacteria with high-producing acidic xylanase activity were screened by transparent zone method, and its fermenta- tion conditions were optimized by combining with single factor experiment and orthogonal experiment. [ Result] A strain B26 of high-producing acidic xylanase was screened and its optimal fermentation medium consisted of: 2% wheat bran, 2% straw powder, 0.8% KNO3 and 0.6% TW-80, amd xylanase activity achieved 1 051.25 U. [ Conclusion] The strain used agricultural waste straw and wheat bran as raw materials to produce high-activity acidic xylanase, and had an important significance for the deep processing, development and utilization of agricultural products.
