摘 要:采用浸浴法,研究低分子质量壳聚糖(LMWC)对镉胁迫引起河南华溪蟹(Sinopotamon henanense)肝胰腺组织中镉积累情况、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量、谷胱甘肽硫转移酶(GST)活性和还原型谷胱甘肽(GSH)含量的影响,旨在评价LMWC的保护作用。设置镉处理组(29、58、87mg/L)、LMWC组(20、40、80mg/L)、LMWC组(20、40、80mg/L)和镉(58mg/L)联合处理组,对照组;处理时间为96h。结果显示:与对照组相比,LMWC单独作用下,SOD活性和MDA含量变化不显著,不会引起肝胰腺组织脂质过氧化。但58mg/L镉单独作用下,SOD活性显著降低,MDA含量显著增加,降低了机体抗氧化能力。镉积累量随着镉浓度升高逐渐增加,加入LMWC后镉积累量显著降低,且具有质量浓度-效应关系。GST活性和GSH含量随着镉质量浓度升高逐渐降低,而加入LMWC后其水平比镉染毒组(58mg/L)显著升高。LMWC对镉胁迫引起的肝胰腺组织氧化损伤可以起到良好的保护作用。To investigate the protective effect of low molecular weight chitosan (LMWC) against cadmium-induced toxicological activity, the freshwater crab Sinopotamon henanense was challenged with LMWC (20, 40 mg/L and 80 mg/L) and or cadmium (29, 58 mg/L and 87 mg/L). Cadmium accumulation, superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, glutathione S-transferase (GST) activity and glutathione (GSH) content were measured in hepatopancreas of S. henanense. The results showed that Cd〉 (58 mg/L) significantly decreased SOD activity and increased MDA content in hepatopancreas of the crabs compared to the control group (P 〈 0.05), whereas LMWC did not change SOD activity and MDA content in hepatopancreas. Also, there was an apparent accumulation of Cd^2+ in hepatopancreas, which was lessened in the presence of LMWC in a concentration-dependent manner. In addition, Cd^2+ significantly decreased the hepatopancreas GST activity and GSH content in the crabs relative to the control (P 〈 0.05). Co-treatment with LMWC enhanced the GST activity and GSH content in hepatopancreas tissues compare with the crabs exposed to Cd^2+ alone. According to these results, it can be concluded that LMWC has protective function against the adverse influence of cadmium on Sinopotamon henanense.
