摘 要:为了提高丁酸的产量,采用响应面分析法对嗜热丁酸梭菌厌氧发酵条件进行优化。在单因素试验的基础上,采用Box-Behnken试验设计,选定培养温度、pH和接种量3个关键因子为响应因子,以丁酸产量为响应值建立多元二次回归方程,在分析各个因素的显著性和交互作用后,确定嗜热丁酸梭菌发酵产酸的最优条件为:培养温度为56.08℃、pH为6.61、接种量的体积分数为6.10%,丁酸产量最大预测值为20.69 g/L。试验验证实际产量为20.36 g/L,比未优化前产量提高了64.8%。To increase the yield of butyric acid, the fermentation conditions were optimized by single factor and response sur- face methodology (RSM). Based on single factor experiment, Box-Behnken center-composite experimental design used, three critical factors (temperature, pH and inoculums size) were selected as response factors, and the butyric acid yield was used as response value to establish the quadric regression equation. Through the analysis of the significance and the interaction of each factor, the optimum fermentation conditions of butyric acid production were as follows; temperature of 56.08 ℃, pH of 6.61, and inoculums size of 6.10% (V/V). Under the optimal conditions, the predicted yield of fitted model was 20.69 g/L. The results verification test was 20.36 g/L, which increased 64.8% compared with the original conditions.
