摘 要:目的研究Wnt基因家族中Wnt1和Wnt信号通路抑制因子DKK1在宫颈鳞癌中的表达及意义。方法应用免疫组织化学法检测宫颈鳞癌及正常宫颈组织中Wnt1和DKK1蛋白的表达。结果宫颈鳞癌中Wnt1的阳性表达率为71.67%,显著高于正常宫颈组织中的40.00%(P<0.05)。DKK1在宫颈鳞癌中的阳性表达率为43.33%,显著低于正常宫颈组织中的66.67%(P<0.05)。在宫颈鳞癌组织中,Wnt1和DKK1与FIGO分期、组织分化程度、有无淋巴转移及浸润深度有关(P均<0.05)。Wnt1与DKK1在宫颈鳞癌中的表达呈负相关(P<0.01)。结论在宫颈鳞癌中,Wnt1作为Wnt信号通路的始动因子发挥作用,而DKK1作为Wnt信号通路的抑制因子发挥作用。二者有望成为宫颈鳞癌判断预后及指导临床治疗的监测指标。Objective To study the significance of expression of Wnt1 and DKK1 in patients with cervical squamous cell carcinoma.Methods Immunohistochemical method was used to detect the protein expression of Wnt1 and DKK1 in specimens of cervical squamous cell carcinoma and normal cervix.Results The positive rate of expression of Wnt1 in SCC(71.67%) was significantly higher than that in normal cervix(40.00%).The positive rate of DKK1(43.33%) in SCC was significantly lower than that in normal cervix(66.67%).The expression of Wnt1 and DKK1 in SCC was associated with clinical staging,differentiation,depth of invasion and metastasis in lymph nodes(P 0.05).The expression of DKK1 was negatively correlated with that of Wnt1(P 0.01).Conclusion DKK1 is an inhibitive factor in signaling pathway of Wnt / β-catenin,but Wnt1 is an initiative factor of this pathway in SCC.They can be regarded as clinical assistant reference markers and offer some evidence for prognosis of SCC.
