摘 要:目的通过检测不同口腔念珠菌的细胞外代谢产物,绘制不同念珠菌的特征性代谢谱图,探讨基于核磁共振的代谢组学方法在快速鉴别不同念珠菌的种类和分析相关代谢产物的化学物质种类的可能性。方法分别在沙保培养基中接种白色念珠菌、克柔念珠菌、光滑念珠菌、热带念珠菌,采取比浊法计数,绘制生长曲线,取生长活跃期的四种念珠菌培养液,行核磁共振检测和主成分分析,比对webspectra数据库中的化学物质种类的标准谱图,得出相关代谢组分。结果四种念珠菌的细胞外代谢产物具有各自的类聚关系,主成分分析法可以区分这四种念珠菌;四种念珠菌的代谢组化学物质种类不同,可能与不同念珠菌菌种有关。结论基于核磁共振的代谢组学检测方法在口腔念珠菌的快速检测中和相关代谢产物的分析中有很好的应用前景。Objective To study the characteristic metabolic profile of Candida albicaus, Candida glabrata, Candida krusei and Candida tropicalis through detecting the extracellular metabolite based on 1H-NMR, so as to quickly identify each of oral Candidas and analyze the extracellular metabofite of different Candidas. Methods The foregoing four Candidal species were cultured respectively into Sabouraud medium. The nephelometry count and growth curve was determined periodically. Each of four Candidas culture medium in the active period of growth was laid under 1H-NMR detection, and principal component analysis (PCA) was conducted.Relevant metabolic components were finally drawn by comparing with Webspectra database. Results The four Candida extracellular metabolites had a respective class of clustering relationship while the PCA analysis could differentiate among the four kinds of Candidas. The four Candida metabolomics had different chemical substances which may be associated with different Candida strains. Conclusion The metabonomics detection method based on 1H-NMR is a potential classable method to quickly identify the oral Candidas. The related metabolites analysis also had a good application prospect.
