摘  要:应用液相色谱-串联质谱技术研究了氟苯尼考在中华鳖日本品系(Pelodiscus sinensis Japanesestrain)体内的残留代谢规律。在(25.0±2.0)℃水温下,150只体重为(250±50)g健康中华鳖日本品系连续7 d投喂含30 mg/kg氟苯尼考的饲料,分别于最后一次给药后1 h、2 h、4 h、8 h、16 h、24 h、48 h、72 h、96 h、120 h、144 h、168 h、240 h、360 h采集肝、血液、肌肉和肾样品。样品中氟苯尼考的残留量采用乙酸乙酯提取,正己烷净化,电喷雾负离子多反应监测模式下进行测定,内标法定量,并利用3P97药动学软件进行数据分析。结果表明,氟苯尼考在中华鳖日本品系血液、肌肉、肝和肾中达峰浓度分别为167μg/kg、188μg/kg、67.15μg/kg和85.71μg/kg,达峰时间分别为4 h、8 h、8 h和8 h,消除半衰期分别为14.9 h、9.4 h、29.5 h和13.0 h,药动学方程分别为:C血液=210.332e-0.070 6 t+938.161e-0.046 5 t,C肌肉=5 642.635e-0.077 1 t+5 765.891e-0.074 4 t,C肝=111.596e-0.036 0 t+4.339e-0.023 5 t和C肾=176.509e-0.065 4 t+615.697e-0.053 6 t。说明氟苯尼考在中华鳖日本品系体内残留代谢较快,建议休药期为15 d。Pharmacokinetics of florfenicol were studied in tissues from Pelodiscus sinensis Japanese strain using high performance liquid chromatography-tandem mass spectrometry. One hundred fifty healthy soft-shell turtles with the weight of 250 ± 50 g were adnubustered florfenicol at a dosage of 30 mg/kg for 7 consecutive days under the temperature of 25.0±2. 0℃. After the last feeding, tissues including blood, muscle, liver and kidney were collected at 1 h, 2 h, 4 h, 8 h, 16 h,24 h, 48 h, 72 h, 96 h, 120 h, 144 h, 168 h, 240 h and 360 h, respectively. Florfenieols in different tissues were extracted with ethyl acetate, cleaned by hexane, and detected by HPLC-MS/MS with negative electrospray ionization under multiple reaction monitoring. Quantitative analysis was used by internal standard method. Results showed that: The peak times of concentrations of florfenicol in plasma, muscle, liver and kidney were 4 h, 8 h, 8 h and 8 h, with the maximum content of 167μg,/kg, 188 μg,/kg, 67. 15 μg/kg and 85.71 μg/kg, respectively. The elimination half life wasl4.9 h, 9.4 h, 29.5 h and 13.0 h.The concentration-time curves of florfenicol in blood, liver, muscle and kidney were: Cblood=210.332e^(-0.0706)+938.161e^(-0.0465), Cmescle=5642.635e^(-0.0771)+5765.891e^(-0.0744t), Cliver=111.596e^(-0.0360t)+4.339e^(-0.0235t) and Ckidney=176.509e^(-0.0654t)+615.697e^(-0.0536t), respectively. Florfenicol was absorbed and eliminated fast in P. sinensis Japanese strain. The withdraw period should not be less than 15 d under this experiment condition.
