摘 要:目的:探讨EphB4受体及其配体EphrinB2在食管鳞癌组织中的表达及其与血管生成的关系。方法:应用免疫组织化学Envision plus法检测60例食管鳞癌和10例癌旁组织中EphB4受体及其配体EphrinB2的表达和VEGF的表达,采用逆转录多聚酶链反应(RT-PCR)法检测60例食管鳞癌和10例癌旁食管组织中EphB4、EphrinB2和VEGF mRNA的相对含量,并分析EphB4、EphrinB2和VEGF的表达的相关性。结果:食管鳞癌组织中EphB4、EphrinB2、VEGF mRNA和蛋白的表达均高于癌旁正常食管黏膜,差异有统计学意义(P<0.05)。EphB4 mRNA和EphrinB2 mRNA表达正相关(r=0.541,P<0.05),EphB4 mRNA、EphrinB2 mRNA均和VEGF mRNA表达正相关(r=0.642,r=0.582;P均<0.05)。结论:EphB4和EphrinB2在食管鳞癌中高表达,与血管生成密切相关。Objective: This study aimed to investigate the expression of EphB4 receptor and its ligand EphrinB2 in esophageal squamous cell carcinoma and their relationship with angiogenesis. Methods: The expression of EphB4 receptor and its ligand EphrinB2 and VEGF was detected by immunohistochemistry in tumor specimens from 60 esophageal squamous cell carcinoma cases and paratumor normal specimens from l0 cases. The expression of EphB4, EphrinB2, and VEGF mRNA in 60 patients with esophageal cancer and l0 patients with paratumor normal esophageal mucosa was detected by reverse-transcription polymerase chain reaction. The correlation among the expression levels of EphB4, EphrinB2, and VEGF mRNA was analyzed. Results: The expression levels of EphB4, EphrinB2, and VEGF mRNA, as well as of the expression levels protein, were significantly higher than that in paratumor normal esophageal mucosa (P〈0.05). The relative amount of EphB4 mRNA was positively correlated with EphrinB2 mRNA in patients with esophageal cancer (r=0.541, P〈0.05). The relative amounts of EphB4 mRNA and EphrinB2 mRNA were positively correlated with VEGF mRNA in patients with esophageal cancer (r=0.642 and 0.582, respectively; P〈0.05). Conclusion: EphB4 and EphrinB2 are highly expressed in esophageal cancer tissue, which may be closely associated with the angiogenesis of esophageal cancer.
