CXCR3阳性自然杀伤细胞在MHV-3诱导暴发性肝炎小鼠组织分布动态变化

摘　　要：目的检测CXC趋化因子受体3(CXCR3)阳性自然杀伤(NK)细胞在3型鼠肝炎病毒诱导的暴发性肝炎小鼠肝脏、脾脏、骨髓及外周血中百分比的动态变化。方法腹腔注射3型鼠肝炎病毒诱导BALB/cJ小鼠暴发性肝炎,利用流式细胞术检测小鼠肝脏、骨髓、外周血及脾脏CXCR3阳性NK细胞百分比的变化。结果肝脏CX-CR3阳性NK细胞百分比在感染后12小时(20.47±1.54%)和24小时(31.53±2.53%)较感染前(14.0±0.25%)均显著升高(P均<0.05),至感染48小时下降至感染前水平;骨髓CXCR3阳性NK细胞百分比随感染时间延长而逐渐减少,由初始水平(25.53±3.50%)的高点至72小时时降至较低水平(1.2±0.17%,P<0.05);脾脏CXCR3阳性NK细胞的百分比在感染后12小时增高(13.63±3.167%),随后下降,至72小时达到较低水平(2.20±0.53%,P<0.05);外周血CXCR3阳性NK细胞的百分比在24小时增高,在72小时下降至感染前水平。结论在MHV-3感染后,CXCR3阳性NK细胞可能逐渐迁移至肝脏,从而在小鼠暴发性肝炎发生发展中发挥重要作用。Objective To investigate the dynamic changes of CXCR3-positive nature killer cells（NK cells） in the liver,the spleen,the bone marrow（BM） and the peripheral blood in a murine fulminant hepatitis model induced by murine hepatitis virus type 3（MHV-3） infection.Methods MHV-3-induced fulminant hepatitis was established in mice,and the percentage of CXCR3-positive NK cells in the liver,the BM,the peripheral blood and the spleen was analyzed by flow cytometry.Results Comparing with the baseline（14.0±0.25%）,the percentage of CXCR3-positiveNK cells was significantly increased at 12h（20.47±1.54%） and 24h（31.53±2.53%） in the liver and decreased to the baseline at 48h;in the bone marrow,CXCR3-positive NK cells decreased gradually,from 25.53±3.50% at baseline to 1.2±0.17% at 72h（P0.05）;in the spleen,CXCR3-positive NK cells was increased at 12h（13.63±3.167%）,and decreased gradually to 2.20±0.53% at 72h;in the peripheral blood,CXCR3-positive NK cells reached the highest level at 24h,but then decreased to the baseline at 72h.Conclusion These findings suggest that the recruitment of CXCR3-positive NK cells into the liver may play an important role in the pathogenesis of murine fulminant viral hepatitis induced by MHV-3 infection.
