摘  要:【目的】观察腹部不同手术切口刺激对急性应激反应晚期心肌组织热休克蛋白70(HSP70)和高迁移率族蛋白昏1(HMG13-1)表达的影响。【方法】健康成年雄性SD大鼠42只,随机分为7组(n=6),C组为对照组,丙泊酚镇静后不作任何手术切口。S2组至E2组为实验组,丙泊酚镇静后依分组的不同分别作不同长度和深度的腹部正中手术切口。7d后处死大鼠,取心肌组织标本,检测HSP70和HMGB-1在心肌中的表达并测定其IOD值。【结果】各组大鼠心肌组织HSP70主要定位于细胞浆中,S2组至E2组大鼠心肌组织HSP70的表达程度随手术刺激强度的加强而有递增趋势,但无统计学意义(P〉0.05);各组大鼠心肌组织HMG13-1的表达主要定位于细胞核,S2组至E2组大鼠心肌组织HMGB-1表达程度随手术刺激强度的加强而递增,且差异有显著性(P〈0.01)。【结论】急性应激反应晚期心肌组织HMG13-1的表达明显增加,而且随手术刺激强度的加强而递增;而心肌组织HSP70的表达无明显增加。[Abstraet][Objective]To observe the effect of different abdominal incision stimulation on the expression of heat shock protein 70(HSP70) and high mobility group protein-l(HMGB-1) in myocardial tissues of SD rats at the late stage of acute stress reaction. [Methods]Totally 42 adult male SD rats were randomly divided into 7 groups with 6 in each. Group C was control group. No incision in group C after propofol anesthesia was made. Groups S2-E2 were experimental groups. According to the different groups, different length and depth of abdominal midline incisions were made in groups S2-E2 after propofol anesthesia. Rats were sacrificed after 7d. Myocardial specimens were prepared. The expression of HSP70 and HMGB-1 in myocardium was detected and IOD was measured. [ResultslThe expression of HSP70 was mainly in cytoplasm of myocardial tissues in all groups. The expression of HSP70 in myocardial tissues of rats in groups S2-E2 showed an increasing trend with the elevation of incision stimulation intensity, but there was no significant difference( P〉0.05). The expression of HMGB-1 was mainly in the nucleus of myocardial tissues in rats of each group. The expression of HMGB-1 in myocardial tissues of rats in groups S2-E2 gradually increased with incision stimulation intensity, and there was significant difference( P 〈0.01). [Conclusion] The expression of HMGB-1 in myocardial tis- sues increases obviously in the late stage of acute stress reaction, and gradually rises up with incision stimula- tion intensity. But the expression of HSP70 in myocardial tissue has no obvious elevation. [Key words] Abdomen/SU; Myocardium; HSP70 Heat-Shock Proteins; HMGB1 Protein
