摘  要:目的 探讨胶质瘤细胞血小板源生长因子 (PDGF)BB表达水平与肿瘤间质血管(SBV)增生的关系。方法 用原位杂交、免疫组化和HE染色观察了 73例人胶质瘤。结果  73例中 ,仅 8例 ( 11% )有少许SBV表达PDGFBmRNA ,SBV的PDGFα受体、β受体和酪氨酸磷酸化蛋白 (P Tyr)的阳性率分别为 95 9%、 34 3%和 6 1 6 % ,四者间均有显著性差异 (P <0 0 0 5 )。 6 2例( 84 9% )的肿瘤细胞表达PDGFBmRNA ,其表达水平与SBV的密度及P -Tyr阳性率均呈平行递增 ,三者也均随肿瘤恶性等级升高而相应增加 ( P <0 0 5~ 0 0 0 1)。结论 本组结果提示胶质瘤细胞普遍表达PDGFBB 。Objective To investigate the relationship between paracrine effect of plateletderived growth factor(PDGF) BB and reactive hyperplasia of stromal blood vessels(SBVs) in human gliomas. Methods Seventythree tissue specimens of human glioma were studied by in situ hybridization, immunohistochemistry and HE staining techniques.Results SBVs expressing PDGFB mRNA were observed only in 8 (110%) of 73 cases. The positive rates of PDGF α receptor, PDGF β receptor and phosphotyrosine protein (PTyr) of the SBVs were 959%(70/73), 343%(25/73) and 616%(45/73) respectively. There was significant difference among above four targets(P<0005). The tumor cells were verified to express PDGFB mRNA in 62 (849%) of 73 gliomas. The expressive level of PDGFB mRNA in glioma cells increased parallely with PTyr positive rate and density of the SBVs as well as all of them were elevated with increase of malignant degree of the tumors (P<005~0001). Conclusion Above data suggest that PDGFBB secreted by glioma cells can stimulate hyperplasia of SBVs in paracrine fashion, PDGF receptors expressed in SBVs and activated by PDGFBB are mainly PDGF α receptor, PDGFBB in autocrine fashion do not play a major role in mediating hyperplasia of SBVs.
